摘要
目的:确定黔产毛茛总皂苷的最佳提取方法和工艺。方法:以黔产毛茛中总皂苷含量为评价指标,考察水、甲醇、95%乙醇三种溶媒分别采用超声、回流及微波三种提取方法对总皂苷提取率的影响,优化提取方法。单因素分析法确定乙醇体积分数、液固比和提取时间为影响因素,采用正交试验设计法安排实验,利用直观分析法结合方差分析法优选最佳提取方法的工艺参数。结果:以85%乙醇为溶媒的回流法提取效果最佳,最优工艺参数为:提取温度80℃,溶媒用量10倍,提取2次,每次2h。结论:本方法测定结果准确、重复性好、专属性强,可为黔产毛茛药材质量控制提供理论依据。
Objective To optimize the extract technology of selecting total saponins from RanuncuLus japonicus T. Methods To optimized extraction method of using the content of total saponins in RanuncuLus japonicus T. in the South of Guihou province as the evaluation index,the effects of three extraction methods of water, methanol and 95% ethanol on the extraction rate of total saponins were studied.The volume fraction, liquid-solid ratio and extraction time of ethanol were determined by the single factor analysis method as the influencing factors, and the experiments were arranged by the orthogonal experimental design method, and the process parameters of the optimal extraction method were optimized by the combination of the intuitive analysis method and the variance analysis method. Result Ten times quantity of 85% ethanol was extracted twice (2h each time) at 80 ℃ as the best extract plan. Conclusions This method is accurate, reproducible and specific, and can provide a theoretical basis for quality control of RanuncuLus japonicus T. in the South of Guizhou.
