改进型胶原蛋白海绵填塞拔牙窝后止血和促愈合的小鼠实验研究 被引量:7

摘要 目的观察改进型胶原蛋白即刻植入小鼠拔牙窝后对拔牙术后止血、材料降解以及拔牙窝愈合效果的影响。方法①随机表法将48只雄性C57BL/6J小鼠分为三组,每组16只,腹腔注射1. 5%戊巴比妥溶液麻醉后,拔除右上颌中切牙,实验组植入改进型胶原蛋白海绵,对照组植入传统型胶原蛋白海绵,空白对照组无特殊处理;②通过沾取三组拔牙后创口渗血,称重并比较三组术后出血量;③于术后第1、2、4、6周,观察三组大体拔牙伤口愈合情况,同时取上颌骨组织,常规组织学切片,Masson染色,观察拔牙窝内胶原蛋白海绵降解、残留植入物大小、植入部位组织学反应、炎性反应、周围组织长入和修复情况。结果改进型胶原蛋白海绵组出血量为0. 010863±0. 007085 g,低于空白对照组0. 038513±0. 022848 g(P <0. 05),传统型止血海绵组出血量为0. 017025±0. 008925 g,也低于空白对照组(P <0. 05);而实验组和对照组出血量差异无统计学意义。组织学观察发现,不同胶原蛋白海绵单独植入拔牙窝后促进拔牙创早期于愈合和后期的骨量形成的效果都优于空白对照,改进型蛋白胶原海绵组拔牙后胶原的降解较快、骨形成的速度优于传统型胶原蛋白海绵组。结论改进型胶原蛋白海绵具有良好的止血效果和更好的促进拔牙创口的愈合和骨形成的作用。 Objective To observe the effect of modified hemostatic collagen on hemostasis,material degradation and healing of extraction socket in mice. Methods 48 male C57 BL/6 J mice were divided into three groups,following the table of random number,16 mice in each group.After intraperitoneal injection of 1. 5% Pentobarbital anesthesia,the right maxillary central incisor was extracted minimally. The experimental group was implanted with modified collagen,while the control group was implanted with traditional hemostatic sponge,while the blank control group had no special treatment. After extraction of teeth,wound bleeding was evaluate by weighing and comparing the amount of bleeding after operation in the three groups. At the first,second,fourth and sixth weeks after operation,the healing of the extraction wounds was observed in three groups. The maxillary of the three groups were dissection at the first,second,fourth and sixth weeks after operation. The collagen sponge degradation,the histological reaction,such as the inflammatory reaction,the remolding of bone tissues and the restoration were observed by routine histological sections and Masson staining. Results The bleeding volume of the experimental group( 0. 010863 ± 0. 007085 g) was lower than that of the blank control group( 0. 038513 ± 0. 022848 g)( P < 0. 05). The bleeding volume of the control group( 0. 017025 ± 0. 008925 g) was also lower than that of the blank control group( P < 0. 05). There was no significant difference between the two sponge groups( P > 0. 05). Histological observation showed that the two groups of collagen in the extraction socket could promote the early healing of the extraction wound and bone formation in the later stage. The collagen degradation of the modified collagen and the bone formation rate was faster than that of the traditional collagen sponge. Conclusion The modified collagen sponge has good hemostatic effect and can promote the healing and bone formation in the extraction sock.
