摘要 目的探讨肿瘤坏死因子超家族成员15(TNFSF15)启动子区遗传变异与肝癌遗传易感性的关系。方法入选300例肝癌患者和430例健康对照。用聚合酶链反应限制性片段长度多态性(PCR-RFLP)技术对TNFSF15-358T>C(rs6478109)和-638A>G(rs7848647)进行基因分型。用非条件Logistic回归分析TNFSF15基因遗传变异与肝癌发病风险之间的关系。结果与TNFSF15-358TT基因型携带者相比,CC基因型携带者肝癌的发病风险增加(OR=1.95,95%CI=1.29~2.95,P<0.01)。性别和年龄分层分析显示,-358CC基因型可影响男性和高龄人群肝癌发病风险,其OR(95%CI)值分别为1.74(1.09~2.77)和2.69(1.42~5.09)。吸烟分层分析显示,-358T>C变异在不吸烟组与吸烟组均与肝癌的易感性相关,OR(95%CI)分别为1.79(1.02~3.13),2.63(1.02~3.68)。TNFSF15-638A>G遗传变异和肝癌的发病风险无关(P>0.05)。结论TNFSF15-358T>C遗传变异影响肝癌的遗传易感性。 Objective To investigate the correlations between tumor necrosis factor superfamily15(TNFSF15) polymorphisms and genetic susceptibility to liver cancer. Methods This case-control study includes 300 liver cancer patients and 400 health controls. Polymorphisms of-358 T>C(rs6478109) and-638 A>G(rs7848647) were detected by polymerase chain reaction restriction fragment length polymorphism(PCR-RFLP). Unconditional Logistic regression was used to analyze the correlations between the genetic variation of TNFSF15 gene and the risk of liver cancer. Results Compared with-358 TT genotype car-riers,-358 CC genotype carriers had an increased risk of liver cancer(OR=1.95, 95% CI=1.29-2.95, P<0.01).Gender and age stratification analysis showed that the-358 CC genotype could affect the risk of liver cancer in males and elderly people with OR(95% CI) values of 1.74(1.09-2.77) and 2.69(1.42-5.09), respectively. Smoking stratification analysis indicates that-358 T>C genotype carriers were associated with susceptibility to liver cancer in both non-smoker and smoker groups;OR(95% CI) were 1.79(1.02-3.13) and 2.63(1.02-3.68). Results also showed that TNFSF15-638 A>G genetic variation was not associated with the risk of liver cancer(P>0.05).Conclusion The genetic variation of TNFSF15-358 T > C is associated with the genetic susceptibility of liver cancer.
