藏山羊AQP7基因克隆和不同组织器官差异表达分析 被引量:3

摘要 [目的]本研究旨在克隆获得藏山羊AQP7基因序列,并构建组织表达谱。[方法]利用RT-PCR技术克隆藏山羊AQP7基因序列,利用相关生物学软件进行生物信息学分析,同时利用qPCR检测AQP7在藏山羊各组织中的表达差异。[结果]克隆获得藏山羊AQP7基因序列长度为1119 bp,其中CDS区为993 bp,编码330个氨基酸。组织表达谱显示AQP7在藏山羊各组织中均有表达,其中在脂肪和肾脏组织中表达量最高,极显著高于其他组织(P<0.01)。[结论]本试验研究结果为藏山羊肌内脂肪研究提供了分子理论基础。 [Objective]The aim of this study was to clone and obtain the AQP7 gene sequence of Tibetan goats,and to construct tissue expression profiles.[Method]The AQP7 gene sequence of Tibetan goats was cloned by RT-PCR and bioinformatics analysis was performed through relevant biological software.Quantitative real-time PCR(qPCR)was used to detect the expression differences of AQP7 in Tibetan goats.[Result]The length of the AQP7 gene sequence was 1119 by,and the length of the CDS region was 993 by,encoding 330 amino acids.The tissue expression profiling showed that AQP7 was expressed in all tissues of Tibetan goats,with the highest expression in Adipose Tissue and kidney Tissues,and significantly higher than other tissues(P<0.01).[Conclusion]The results of this study provide molecular theoretical basis for the study of intramuscular fat in Tibetan goats.
