基于网络药理学探究泻白糖浆治疗咳嗽的作用机制

Network pharmacology study on mechanism of Xiebai Syrup for cough

目的筛选泻白糖浆的活性成分,寻找其治疗咳嗽的核心靶点,探讨其治疗咳嗽的作用机制。方法采用网络药理学方法。结合文献和中药系统药理学数据库和分析平台(Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform,TCMSP)查询并筛选泻白糖浆的活性成分,通过Swiss Target Prediction数据库和STITCH数据库进行活性成分作用靶点的预测,采用Therapeutic Target Database(TTD)数据库、Genetic Association Database(GAD)数据库、遗传药理学与药物基因组学数据库(Pharmacogenetics and Pharmacogenomics Knowledge Base,PharmGKB)寻找与咳嗽相关的靶点基因,并通过Uniprot数据库进行标准化。采用Cytoscape 3.7.1软件构建蛋白质相互作用网络,寻找核心靶点。采用Metascape数据库对核心靶点进行基因本体通路富集分析。结果筛选得到核心靶点54个,找出靶点的相关通路20条,作用通路涉及内质网应激反应和免疫反应激活信号转导等生物过程。结论本研究体现了中药多成分-多靶点-多途径的作用特点,为泻白糖浆的临床用药提供了一定的理论依据。

Objective To screen the active ingredients of Xiebai Syrup and to explore its core targets and mechanism for the treatment of cough.Methods Network pharmacology methods were employed.The active ingredients of the Xiebai Syrup were found and screened by Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform(TCMSP)and literature.Gene targets related to cough were searched using the Therapeutic Target Database(TTD),the Genetic Association Database(GAD),and the Pharmacogenetics and Pharmacogenomics Knowledge Base(PharmGKB)database and normalized by the Uniprot database.Protein interaction networks was constructed with Cytoscape 3.7.1 software in order to find core targets.The Metascape database was used to analyze Gene Ontology of the core targets.Results Fifty-four core targets were screened and 20 main pathways related with cough were found,involving response to endoplasmic reticulum stress,immune response-activating signal transduction and so on.Conclusion This study reflected the“multi-component,multi-target,multi-pathway”characteristics of Xiebai Syrup,which provides a theoretical basis of the clinical use of Xiebai Syrup.
