超声波辅助提取绣球菌麦角甾醇的工艺优化 被引量:3

摘要 采用超声波辅助提取绣球菌麦角甾醇的方法,通过单因素试验和正交试验,对影响绣球菌麦角甾醇提取效果的主要因素提取溶剂、料液比和超声波处理时间进行研究,并建立了绣球菌中麦角甾醇紫外分光光度法(UV法)快速检测方法。结果表明,对绣球菌麦角甾醇提取量影响的因素按先后顺序依次为提取剂、超声波提取时间和料液比;最佳提取工艺条件为以三氯甲烷为提取剂、料液比1∶20(g/mL)、超声波提取时间80min,在此条件下,得到的麦角甾醇含量最高。UV法测定麦角甾醇的线性范围为5.00~50.00μg/mL,相关系数为0.9945,该方法的精密度、稳定性和重复性都表现良好,适用于绣球菌生产及对生物量的间接测定。 To optimize the ultrasonic-assisted extration process of ergosterol from the fruiting bodies of Sparassis crispa, influence of solvent types, ration of material-liquid and ultrasonic time were investigated. On the bassis of single factor experiment, the extration process was optimized by orthogonal tests. And a rapid quantitative determination of ergosterol in Sparassis crispa through UV method was established. The results showed that the factors affecting the extraction amount of ergosterol were in the order of solvent types, ultrasonic time and ratio of material-liquid. The optimal extration conditions was chosen as trichloromethane, 1∶20(g/mL) and 80 min for solvent types, ration of material-liquid and ultrasonic time, respectively, the highest ergosterol content was obtained under these conditions. The liner ranges of ergosterol through UV method were 5.0-50.00 μg/mL, correlation coefficient was 0.994 5. This method showed good precision, stability and repeatability, which was suitable for the production of ergosterol and indirect determination of biomass of Sparassis crispa.
