摘要
HAB-9是从槟榔树根际土壤中分离到的一株解淀粉芽孢杆菌(Bacillus amyloliquefaciens),该菌株对17株植物病原真菌有较好的拮抗作用。经16srDNA及生化特征测定,鉴定HAB-9菌株,应用对峙法测定了抑菌效果。结果显示,经16S rDNA鉴定,HAB-9菌株为B.amyloliquefacien;经生化特征测定,其革兰氏染色为阳性,M.R测定呈阴性,V-P测定为阳性,氧化酶测定为阴性,接触酶测定为阳性,淀粉水解试验发现具有解淀粉的功能;PDA平板对峙培养,发现HAB-9菌株对槟榔炭疽菌(Colletotrichum gloeosporioides)等17株植物病原真菌的抑菌活性有较大差异,平均抑制率为60.82%;对槟榔炭疽菌(C.gloeosporioides)有较好的拮抗作用,其抑菌率为70.59%;效果最差的为黄瓜炭疽病菌(C.orbiculare),抑菌率为50.22%。从本实验结果可知,HAB-9菌株对多数病原炭疽菌有较强的抑制作用,具有潜在的应用价值。
HAB-9 is a strain of Bacillus amyloliquefaciens isolated from rhizosphere soil of arecanut.The strain had good antagonistic effect on 17 strains of plant pathogenic fungi.HAB-9 strain was identified by using 16S rDNA and biochemical characteristics.The antifungal effect was then determined by using the confrontation method.The results showed that strain HAB-9 was identified as B.amyloliquefacien by 16SrDNA.It is Gram positive,MR negative,V-P positive,oxidase negative and catalase positive.Starch hydrolysis test showed that it had the function of starch hydrolysis.PDA plate confrontation culture test found that the strain HAB-9 had a significant difference in an antifungal activity against 17 strains of plant pathogenic fungi like Collectotrichum gloeosporioides,with an average inhibition rate of 60.82%.It had a good antagonistic effect on Colletotrichum gloeosporioides,and its fungal inhibition rate was 70.59%.It had the worst inhibition effect on C.orbiculare,with an inhibition rate of 50.22%.According to the results of this experiment,HAB-9 strain has a high inhibitory effect on most of Colletotrichum pathogens,which indicates that it has a potential application value.
