β-catenin和Axin在化生性乳腺癌中的表达及其临床意义 被引量:1

Expression and clinical significance of β-catenin and Axin in metaplastic breast carcinomas

摘要 目的探究化生性乳腺癌(metaplastic breast cancer,MBC)中β-catenin蛋白及Axin蛋白的表达及其与临床病理特征的关系.方法本实验收集了2006年6月至2015年12月在哈尔滨医科大学附属肿瘤医院接受手术且术后经病理证实为MBC的68例患者的乳腺癌组织和22例乳腺癌癌旁正常组织.利用免疫组织化学(immunohistochemistry,IHC)法检测MBC患者的原发灶及癌旁正常组织中β-catenin蛋白及Axin蛋白的表达情况,并利用统计学的方法分析两种蛋白与MBC患者临床病理特征的关系及两种蛋白之间的相关性,同时根据随访资料分析β-catenin蛋白及Axin蛋白表达与MBC复发转移的关系.结果在MBC组织中β-catenin和Axin的阳性表达率分别为45.6%(31/68)和47.1%(32/68).β-catenin的表达与肿瘤大小、临床分期呈正相关(r值分别为0.312和0.351,P值均<0.05).Axin表达与肿瘤大小、临床分期呈负相关(r值分别为-0.378和-0.324,P值均<0.05).以上两种蛋白表达与年龄、淋巴结转移、绝经状态和P53表达均不相关.β-catenin蛋白与Axin蛋白的表达呈负相关(r=-0.39,P=0.001),且均与MBC复发、转移相关(r值分别为0.271和-0.351,P值均<0.05).结论β-catenin与Axin可能是MBC患者潜在的治疗靶点. Objective To investigate the expression of β-catenin and Axin in metaplastic breast cancer (MBC)tissues,and to understand the relationship between their expression and the clinicopathological parame-ters. Methods Total of 68 breast cancer tissues and 22 cases of breast cancer adjacent normal tissues were col-lected from patients who underwent surgery from June 2006 to December 2015 in the Affiliated Tumor Hospital of Harbin Medical University and confirmed by pathology as MBC. Immunohistochemical(IHC)method was used to detect the expression of β-catenin and Axin protein in primary lesions and adjacent normal tissues of pa-tients with MBC. The relationships between the expression of the two protein themselfs(β-catenin protein and Axin protein)and the clinicopathological characteristics of MBC patients were analyzed. In addition,the rela-tionships between the expression of the two protein and the recurrence and metastasis of MBC were also analyzed according to the follow-up data. Results The positive rates of β-catenin and Axin in MBC tissues were 45. 6%(31 / 68)and 47. 1%(32 / 68),respectively. The expression of β-catenin was positively correlated with tumor size and clinical stage(r values were 0. 312 and 0. 351 respectively,both P values < 0. 05). The expression of Axin was negatively correlated with tumor size and clinical stage(r values were - 0. 378 and - 0. 324,both P values < 0. 05). The two proteins were not correlated with age,lymph node metastasis,menopausal status and P53 expression. There was a negative correlation between the expression of β-catenin and Axin(r =- 0. 39,P <0. 05). The expression of β-catenin and Axin were correlated with the recurrence and metastasis of MBC(r val-ues were 0. 271 and - 0. 351 respectively,both P values < 0. 05). Conclusion β-catenin and Axin may be po-tential treatment target for patient with MBC.
