冬瓜皮炭对慢性肾衰竭大鼠肾功能的保护作用 被引量:4

摘要 目的探讨冬瓜皮炭对慢性肾衰竭大鼠肾功能的保护作用。方法 72只大鼠随机分为正常组、模型组、阳性对照组(爱西特,450 mg/kg)及冬瓜皮炭低、中、高剂量组(300、450、600 mg/kg)。腺嘌呤灌胃给药30 d后,ELISA法检测血清BUN、SCr、TGF-β1、PAI-1水平,HE染色、Masson染色观察肾组织病理形态变化,RT-PCR检测NF-κB p65、I-κB mRNA表达,Western blot检测NF-κB p65蛋白表达。结果与模型组比较,冬瓜皮炭高剂量组BUN、SCr、TGF-β1、PAI-1水平,NF-κB p65、I-κB mRNA表达,NF-κB p65蛋白表达显著降低(P<0.05,P<0.01),肾小管轻度扩张、肾小管上皮细胞轻度空泡样变性、肾间质纤维化、炎细胞浸润明显缓解。结论冬瓜皮炭(600 mg/kg)能降低慢性肾功能衰竭大鼠肾脏有害指标,改善肾功能,其机制可能与干预NF-κB信号通路有关。 AIM To explore the protective effects of Benincasae Exocarpium charcoal on renal functions in rats with chronic kidney failure.METHODS Seventy-two rats were randomly divided into normal group, model group, positive control group(aceset, 450 mg/kg) and low-dose, medium-dose, high-dose Benincasae Exocarpium charcoal groups(300, 450, 600 mg/kg). After 30-day intragastric administration of adenine, serum BUN, SCr, TGF-β1 and PAI-1 levels were detected by ELISA, the pathological morphology of renal tissues was observed by HE staining and Masson staining. NF-κB p65 and I-κB mRNA expressions were detected by RT-PCR, NF-κB p65 protein expression was detected by Western blot. RESULTS Compared with the model group, the high-dose Benincasae Exocarpium charcoal group demonstrated significantly decreased BUN, SCr, TGF-β1 and PAI-1 levels, NF-κB p65 and I-κB mRNA expressions, and NF-κB p65 protein expression(P<0.05, P<0.01), with obviously alleviated mild tubular dilatation, mild vacuolar degeneration of renal tubular epithelial cells, renal interstitial fibrosis and decreased inflammatory cell infiltration.CONCLUSION Benincasae Exocarpium charcoal(600 mg/kg) can reduce renal harmful indices and improve renal functions in rats with chronic kidney failure, whose mechanisms may contribute to the intervention of NF-κB signaling pathway.
