摘要
目的探索槲皮素对结直肠癌细胞SW620迁移和侵袭的影响。方法不同浓度(0、25、50、100μmol/L)槲皮素处理SW620细胞后,MTT法检测细胞存活率,Transwell法检测细胞迁移和侵袭,RT-PCR、Western blot分别检测STAT3 mRNA、蛋白表达。结果槲皮素呈剂量和时间依赖性地降低SW620细胞存活率、STAT3 mRNA表达。最佳浓度、处理时间分别为50μmol/L、48 h,细胞迁移和侵袭显著降低(P<0.05)。敲低STAT3,SW620细胞存活率、迁移和侵袭数目明显减少(P<0.05)。STAT3过表达显著逆转槲皮素对细胞存活率、迁移和侵袭的抑制作用(P<0.05)。结论槲皮素可通过调控STAT3表达来抑制SW620细胞迁移和侵袭。
AIM To explore the effects of quercetin on migration and invasion of colorectal cancer SW620 cells.METHODS SW620 cells were treated with different concentrations(0, 25, 50, 100 μmol/L) of quercetin, after which cell viability was detected by MTT assay, cell migration and invasion were detected by Transwell assay, STAT3 mRNA and protein expressions were detected by RT-PCR and Western blot, respectively.RESULTS Quercetin decreased SW620 cell viability and STAT3 mRNA expression in dose-dependent and time-dependent mannerso the optimal concentration and treatment time were determined to be 50 μmol/L and 48 h, repsectively, along with significantly reduced cell migration and invasion(P<0.05);and the cell viability,migration and invasion of SW620 cells were significantly redued when STAT3 was knocked down(P<0.05). The overexpression of STAT3 obviously reversed the inhibitory effects of quercetin on cell viability, migration and invasion(P<0.05). CONCLUSION Quercetin can inhibit the migration and invasion of SW620 cells by regulating STAT3 expression.
