摘要
目的建立肚液散UPLC指纹图谱,评价不同剂量^60Co-γ对3种成分的影响。方法肚液散70%甲醇提取物的分析采用ACQUITY UPLC BEH C18色谱柱(2.1 mm×100 mm,1.7μm);流动相乙腈-水(含0.05%磷酸),梯度洗脱;柱温25℃;体积流量0.18 mL/min;检测波长237 nm。^60Co-γ设定0、4、8 kGy 3个剂量。结果 13批样品指纹图谱中有22个共有峰,相似度均大于0.94。^60Co-γ射线照射后,肚液散大部分化学成分较为稳定。随着辐照剂量增加,甘草苷含有量明显下降(P<0.05),甘草酸和异甘草素在不同辐照剂量下均较稳定。结论该方法稳定可靠,可用于肚液散的质量控制。
AIM To estabilish the UPLC fingerprints of Duye Powder and to evaluate the effects of different doses of ^60Co-γ irradiation on three constituents. METHODS The analysis of methanol extract of Duye Powder was developed on a 25 ℃ thermostatic ACQUITY UPLC BEH C18 column(2.1 mm×100 mm,1.7 μm), with the mobile phase comprising of cetonitrile-water(containing 0.5% phosphoric acid) flowing at 0.18 mL/min in a gradient elution manner, and the detection wavelength was set at 237 nm.Three different doses of ^60Co-γ irradiation were set as 0,4,8 kGy. RESULTS There were twenty-two common peaks in the fingerprints of thirteen batches of samples, with the similarities of more than 0.94. Most of the chemical constituents in Duye Powder were stable after ^60Co-γ irradiation.The content of liquiritin decreased significantly, with in creasing the irradiation dose(P<0.05), and glycyrrhizic acid and isoliquiritigenin were more stable at different irradiation doses. CONCLUSION This stable and reliable method can be used for the quality control of Duye Powder.
