LN在不同繁殖期蓝狐睾丸组织分布比较

摘要 【目的】比较睾丸细胞外基质的重要成分LN在不同繁殖季节蓝狐睾丸中的分布及变化特点.【方法】采用H.E染色法、组织化学法、免疫荧光技术结合IPP形态计量比较分析.【结果】H.E染色表明繁殖间期与繁殖期蓝狐睾丸生精小管内部结构有明显不同,繁殖期蓝狐睾丸生精上皮细胞层数明显多于繁殖间期,精子发育良好,繁殖间期生精小管内无成熟精子.免疫组织化学密度表明,LN在蓝狐不同繁殖期睾丸间质细胞,小血管壁均呈强阳性表达,管周肌样细胞中等表达,阴性对照无表达.免疫荧光表达显示,LN主要表达于间质细胞及管周肌样细胞,平均吸光度统计结果表明,繁殖期较繁殖间期表达显著增高.【结论】蓝狐睾丸LN在生精小管基膜的表达在不同繁殖期变化不明显,但在间质细胞及支持细胞的表达差异显著与季节性繁殖有一定关系,提示LN可能参与间质细胞分泌雄性激素的调节. 【Objective】 To compare the distribution characteristics of LN in the testicles of Alopex lagopus in different breeding seasons.【Method】 Adopt hematoxylin-eosin staining,histological method,immunofluorescence technique and IPP morphometric comparison were used to compare the localization characteristics of LN in A. lagopus testis in different breeding seasons.【Result】 H.E staining showed that the internal structure of spermatogenic tubules in the testis of A. lagopus was significantly different between the non-breeding season and the breeding season. The number of spermatogenic epithelial cells in the testis of A. lagopus was significantly higher during the breeding season. Immunohistochemical density showed that that LN in the interstitial cells and small blood vessel walls were strongly positively expressed,the peritubular myoid cells were moderately expressed,and the negative control group was not expressed in different breeding seasons of A. lagopus. In the interstitial cells and peritubular myoid cells,the mean absorbance in the breeding season was higher than that in the non-breeding season.【Conclusion】 The expression of LN in spermatogenic tubule basement membrane of A. lagopus testicles was not affected by the reproductive period,but the expression difference of interstitial cells was significantly related to the seasonal reproduction,suggesting that LN may be involved in the regulation of the secretion of male hormones by interstitial cells.
