摘要
[目的]利用基因芯片筛选脊柱转移癌患者组织中差异表达的lncRNAs和mRNAs,分析lncRNAs在脊柱转移癌中可能的分子机制。[方法]选取2016年宁夏医科大学总医院就诊经病理证实的3例脊柱转移癌患者和3例正常对照患者。利用LncPathTMHuman Cancer Array(8×15 K,Arraystar)芯片检测患者和正常对照组织的lncRNAs和mRNAs、GO及KEGGpathway,分析差异表达的lncRNAs和mRNAs的功能分布。[结果]脊柱转移癌患者中差异表达的lncRNAs有326条,差异表达的mRNAs有775条。GO分析发现差异表达的mRNAs主要参与细胞增殖调节、生物细胞过程负调控、局灶性粘连、细胞-基质粘连等。Pathway分析发现差异表达的mRNAs主要参与癌症途径、AGERAGE信号通路与糖尿病并发症、细胞因子及其受体相互作用、慢性粒细胞白血病等。[结论]脊柱转移癌患者椎体组织中差异表达的lncRNAs可能与脊柱转移癌的发生和发展密切相关,未来可能成为脊柱转移癌潜在治疗靶点。
[Objective]To screen the differential expressions of lncRNAs and m RNA in the tissues of spinal metastases by gene chip,and to analyze the possible molecular mechanisms of lncRNAs in spinal metastases.[Methods]Three patients with spinal metastases confirmed by pathology in Ningxia Medical University General Hospital in 2016 and three normal controls were selected.LncPathTM Human Cancer Array(8×15 K,Arraystar)chip was used to detect lncRNAs and mRNAs in the metastatic tumor and normal tissues.In addition,GO and KEGG pathway analysis were conducted to show the differences in expressions of lncRNAs and mRNAs.[Results]The spinal metastatic tumor had 326 lncRNA and 775 mRNA in differential expressions compared with the normal tissues.GO analysis found that differentially expressed mRNAs were mainly involved in cell proliferation regulation,negative regulation of biological cell processes,focal adhesions,and cell-matrix adhesions.Pathway analysis found that differentially expressed mRNAs were mainly involved in the cancer pathway,AGE-RAGE signaling pathway and diabetes complications,cytokine and its receptor interaction,chronic myeloid leukemia.[Conclusion]The differential expression of lncRNAs in vertebral tissue of patients with spinal metastases may be closely related to the occurrence and development of spinal metastases,and may become a potential therapeutic target for spinal metastases in the future.
