摘要
目的:观察HMGB1在哮喘大鼠气道炎症中的作用及苗热咳喘胶囊干预的效果。方法:将40只健康雄性SD大鼠随机分为正常组、模型组、地塞米松组及苗药热咳喘胶囊组;模型组、地塞米松组及苗药热咳喘胶囊组均采用腹腔注射1%卵蛋白(OVA)方法建立哮喘模型,第15天开始每次激发前1小时正常组与模型组予生理盐水灌胃,地塞米松组予醋酸地塞米松注射液腹腔注射,苗药热咳喘胶囊组予苗药热咳喘胶囊灌胃;疗程结束后计算肺泡灌洗液(BALF)中嗜酸性粒细胞(EOS)及中性粒细胞(NEU)数量,检测BALF中高迁移率族蛋白B1(HMGB1)含量。结果:正常组肺泡灌洗液中EOS、NEU计数及HMGB1含量均明显低于模型组,差异具有统计学意义(P<0.05);模型组、苗药热咳喘胶囊组、地塞米松组肺泡灌洗液中HMGB1含量明显高于正常组,具有统计学意义(P<0.05);苗药热咳喘胶囊组、地塞米松组肺泡灌洗液中HMGB1含量明显低于模型组,具有统计学意义(P<0.05)。结论:HMGB1与哮喘发生相关,其可能参与了哮喘气道炎症的形成,而且热咳喘胶囊和地塞米松均能抑制HMGB1的表达。
Objective:To observe the effect of high mobility group B1 protein on airway inflammation in asthmatic rats and the effect of miao re kechuan capsule intervention.Methods:Forty healthy male SD rats were randomLy divided into normal group,model group,dexamethasone group and Miao medicine Rekechuan capsule group.Asthma model was established by intraperitoneal injection of One percent ovalbumin in model group,dexamethasone group and Miao medicine Rekechuan capsule group.Normal group and model group were given saline intragastric administration one hour before each challenge on the fifteenth days,and dexamethasone acetate group was given dexamethasone acetate injection.Miao medicine Rekechuan capsule group was given Miao medicine Rekechuan capsule by intraperitoneal injection.After treatment,the number of eosinophils and neutrophils in alveolar lavage fluid was calculated,and the content of high mobility group protein B1 in alveolar lavage fluid was detected.Results:Eosinophil,neutrophil count and high mobility group protein B1 content in alveolar lavage fluid of normal group were significantly lower than those of model group(P<0.05).The content of high mobility group protein B1 in alveolar lavage fluid of model group,miao yao rekechuan capsule group and dexamethasone group was significantly higher than that of normal group(P<0.05).The content of high-mobility group protein B1 in alveolar lavage fluid of the miao medicine rekechuan capsule group and dexamethasone group was significantly lower than that of the model group,with statistical significance(P<0.05).Conclusion:High mobility group protein B1 is associated with asthma,which may be involved in the formation of airway inflammation in asthma,and both Rekechuan capsule and dexamethasone can inhibit the expression of high mobility group protein B1.
