摘要
目的:探讨没食子儿茶素没食子酸酯是否存在促进3T3-L1脂肪细胞吸收葡萄糖的潜力。方法:为测定没食子儿茶素没食子酸酯对脂肪细胞吸收葡萄糖的作用和对前脂肪细胞分化的影响,分别采用了葡萄糖荧光示踪剂检测法和3T3-L1前脂肪细胞分化模型。结果:没食子儿茶素没食子酸酯可提高脂肪细胞在高浓度葡萄糖(30mM)条件下由胰岛素刺激的葡萄糖吸收,促进3T3-L1前脂肪细胞分化为脂肪细胞。结论:没食子儿茶素没食子酸酯可促进脂肪细胞吸收葡萄糖和前脂肪细胞分化,提示没食子儿茶素没食子酸酯可能对降低血糖和改善胰岛素抵抗有一定作用。
Ethanol extract of Camellia Oleifera Abel.decreased blood glucose in type 2 diabetic mice to a certain extent.Therefore, this study explored whether Gallocatechin-3-gallate which isolated from ethanol extract of Camellia Oleifera Abel.possessed therapeutic potential for enhancing glucose uptake in 3 T3-L1 adipocytes.After 3 T3-L1 pre-adipocytes were induced to differentiate into mature adipocytes, mature adipocytes consumed glucose in the external environment.The effects of insulin or insulin analogues on different enzymes and receptors in cells can activate signaling pathways in many cells and nuclei to further promote the absorption, transportation and consumption of glucose around mature adipocytes, and ultimately lead to the decrease of glucose concentration in cell culture media.Therefore, 3 T3-L1 pre-adipocyte line has been widely used in screening hypoglycemic drugs.In addition, it is very important to select a direct, objective and rigorous method to evaluate the hypoglycemic potential of components.To determine effect of Gallocatechin-3-gallate on glucose uptake and differentiation, the fluorescent tracer of glucose 2-[N-(7-nitrobenz-2-oxa-1, 3-diazol-4-yl)amino]-2-deoxy-D-glucose(2-NBDG)uptake assay and 3 T3-L1 pre-adipocytes differentiation model were adopted.Gallocatechin-3-gallate accelerated insulin-stimulated glucose uptake in differentiated 3 T3-L1 adipocytes under high-glucose conditions(30 mM)and enhanced 3 T3-L1 pre-adipocyte differentiation.Gallocatechin-3-gallate increased glucose uptake in adipocytes and improve 3 T3-L1 pre-adipocyte differentiation, therefore, it might contribute to glycemic control and insulin resistance amelioration.
