摘要
目的探讨TNF-a在外伤性增生性玻璃体视网膜病变(PVR)发病中的作用。方法 32只青紫蓝兔随机分为正常组(8只)与模型组(24只),通过对模型组兔眼建立外伤性PVR模型,分别在造模后10、20和30 d行眼底照相;视网膜组织HE染色、TNF-α免疫组化染色;RT-PCR检测视网膜组织中TNF-αmRNA含量的变化。结果成功建立外伤性PVR模型,HE染色及眼底照相可见模型组PVR的各个分级的表现。模型组在造模后10、20和30 d,视网膜组织中TNF-α免疫染色呈强阳性。TNF-a mRNA的相对表达量明显高于正常组(P <0.05)。结论 TNF-α表达水平升高可能在外伤性PVR的发病中起重要作用。
Objective To explore the role of tumor necrosis factor-α(TNF-α) in traumatic proliferative vitreoretinopathy(PVR).Methods A total of 32 Chinchilla rabbits were randomly divided into normal group(n=8) and model group(n=24). Traumatic PVR models of retina were built for the model group. Fundus photography was performed 10, 20 and 30 days after modeling. HE staining was used for retinal tissues and immunohistochemical(IHC) staining was used for TNF-α. The change of TNF-α mRNA expression level was detected with reverse transcription polymerase chain reaction(RT-PCR). Results The traumatic PVR models were successfully established, and the performance of PVR in model group at different levels was observed by HE staining and fundus photography. IHC staining for TNF-α in retinal tissue showed positive in the model group 10, 20 and 30 days after modeling. The relative expression level of TNF-α mRNA in the model group was significantly higher than that of the normal group(P < 0.05).Conclusion The elevation of TNF-α expression level may play an important role in traumatic PVR.
