基于体外抗氧化法优化红芪总黄酮提取工艺 被引量:5

Optimization of the extraction process of total flavonoids in Radix Hedysari based on the antioxidant activities in vitro

摘要 目的优化红芪总黄酮提取工艺。方法采用Box-Behnken响应面法,以总黄酮的得率及其抗氧化能力(DPPH自由基清除率和O2-清除率)为评价指标,考察溶剂浓度、料液比、回流提取时间3个因素对红芪总黄酮提取工艺的影响。结果经DesignExpert8.0.5b软件处理,由多元二次回归方程的结果得出红芪中总黄酮最佳提取工艺:乙醇浓度为60%,回流料液比为1∶12,回流提取时间为1h。在此条件下红芪药材总黄酮浸膏得率为41.33%,DPPH清除率为54.60%,O2-清除率为48.53%,抗氧化能力最佳。结论使用该法能合理优化红芪总黄酮提取工艺,且红芪具有较强的体外抗氧化活性。 Objective To optimize the extraction process of total flavonoids in Radix Hedysari. Methods The response surface methodology was used for the optimization of the extraction process,with the overall yield and antioxidant activity of total flavonoids(the clearance of DPPH and O2- free radicals)as the evaluation index. The effects of solvent concentration,solid-liquid ratio and reflux extraction time on the extraction process of total flavonoids in Radix Hedysari were investigated. Results The data were analyzed by the Design Expert 8.0.5b software. According to the fitted multivariate quadratic regression equation,the optimal process parameters were as follows:the ethanol concentration was 60%,the extraction time was 1 h and the solid-liquid ratio was 1∶12. Under these conditions,the extraction yield of total flavonoids from Radix Hedysari was 41.33%,the DPPH free radical clearance was 54.60%,and the O2- free radical clearance was 48.53%. The best antioxidant effects of the total flavonoids were thus achieved. Conclusion The present conditions optimized the extraction process of total flavonoids in Radix Hedysari,and the total flavonoids prepared by the optimized process showed a strong antioxidative activity in vitro.
