摘要
目的观察山香圆总黄酮对脂多糖诱导的RAW264.7细胞MAPK信号通路的影响。方法采用脂多糖刺激RAW264.7细胞制备炎症模型;CCK-8法测定细胞活力;ELISA法检测TNF-α、IL-1β、IL-6的分泌量;westernblot法检测ERK1/2、JNK和p38蛋白的磷酸化水平。结果低于400μg/mL的山香圆总黄酮对RAW264.7细胞生长无影响;山香圆总黄酮各剂量组均能抑制p-ERK1/2、p-JNK、p-p38蛋白的表达以及TNF-α、IL-1β、IL-6的分泌,中、高剂量组呈显著性抑制(P<0.05,P<0.01)。结论山香圆总黄酮能通过抑制ERK1/2、JNK、p38磷酸化水平,降低TNF-α、IL-1β、IL-6的分泌,发挥抗炎作用。
Objective To observe the effect of total flavones from Turpinia arguta Seem on MAPK signal pathway in the RAW264.7 cells induced by lipopolysaccharide.Methods RAW264.7 cells were induced by LPS to prepare the inflammatory model.Cell Counting Kit-8 test was used to detect cell viability.ELISA was used to detect the secretion level of TNF-α,IL-1β,and IL-6.Western blot was used to detect the phosphorylation of ER K1/2,JNK,and p38 protein.Results The total flavonoids of lower than 400ug/mL had no effect on the growth of RAW264.7 cells,and each dose group of total flavones from the Turpinia arguta Seem could inhibit the phosphorylation of ERK1/2,JNK,and p38 protein and the secretion of TNF-α,IL-1β,and IL-6.The mid dose and high dose group were significant lower than the model group(P<0.05,P<0.01).Conclusion The anti-inflammatory mechanism of total flavonoids from the Turpinia arguta Seem could be related to its inhibiting of JNK,ERK and p38MAPK signaling pathways to reduce the secretion of TNF-α,IL-1βand IL-6.
