旋覆代赭合六君子汤及其含药血清有效成分研究

Explore the active components in Xuanfu-Daizhe combined with Liujunzi Decoction and its blood seurm.

目的基于液质联用技术(LC-MS)建立同时测定中药复方旋覆代赭汤合六君子汤中白术内酯Ⅲ、槲皮素、茯苓酸三种活性成分的方法,并探究其大鼠含药血清中三种成分含量。方法Agilent-C18柱(2.1mm×150mm,5μm)色谱柱分离,流动相为乙腈(A)-0.1%甲酸水溶液(B)(5∶95),进行梯度洗脱,流速为0.3ml/min,柱温为35℃。结果复方试剂供试液中各有效成分的平均浓度:白术内酯Ⅲ4109.12mg·L^-1;槲皮素50.70mg·L^-1;茯苓酸0.666mg·L^-1;含药血清中白术内酯Ⅲ的平均浓度为55.75mg·L^-1。结论该方法可以用以同时检测旋覆代赭汤合六君子汤中药复方及其大鼠含药血清中上述三种有效物质的含量。

Objective To develop a method for simultaneous determination of three active Components of atractylenolideⅢ,quercetin and pachymic acid in traditional Chinese compound called Xuanfu-Daizhe Decoction combined with Liujunzi Decoction based on the technology of LC-MS.And to explore the content of three components in the blood serum in rats.Methods Agilent-C18 column(2.1mm×150 mm,5μm)was used.The mobile phase consisted of acetonitrile(A)and 0.1%aqueous formic acid(B)(5:95)with gradient elution,and the flow rate was 0.3 ml·min^-1.The temperature of column was 35℃.Results The average concentration of the three effective components in the compound reagent solution is 4109.12mg·L^-1,50.70 mg·L^-1,0.666 mg·L^-1.Also,the average content of atractylenolideⅢin the serum containing drugs is 55.75 mg·L^-1.Conclusion This method can be used for simultaneous determination of three active Components in Xuanfu-Daizhe Decoction combined with Liujunzi Decoction as well as the blood serum contaning these drugs at the same time.
