钩藤对脑损伤早期癫痫易感性的影响 被引量:8

Effects of Uncaria rhynchophylla on the susceptibility of early epilepsy after traumatic brain injury

摘要 目的研究中药钩藤(UR)对脑损伤小鼠早期癫痫的影响,初步确定其抑痫的神经机制。方法采取Feeney自由落体法建立小鼠颅脑损伤模型。通过行为学观察各组小鼠强直、阵挛癫痫发作的潜伏期;经脑电图(EEG)记录分析额叶皮层放电活动;采用脑干湿重法检测脑组织含水量变化;采用免疫荧光法计数海马齿状回(DG)区胶质纤维酸性蛋白(GFAP)的阳性细胞数;采用蛋白免疫印迹法检测海马转化生长因子-β2(TGF-β2)蛋白表达水平。结果与模型组相比,UR低、高剂量组均能延长小鼠强直、阵挛发作潜伏期(P<0.05),减少脑部异常放电,减轻脑水肿(P<0.05),降低海马区星形胶质细胞GFAP阳性细胞数和TGF-β2蛋白的表达(P<0.05)。其中,钩藤高剂量组较模型组改善效果更为显著。结论中药钩藤能够降低脑损伤早期癫痫的易感性,并具有神经保护作用,其机制与星形胶质细胞的调控作用有关。 Objective To study the effects of Uncaria rhynchophylla(UR)on early epilepsy of traumatic brain injury in mice,and initially determine inhibition of epilepsy mechanisms.Methods A mouse model of closed brain injury was established by the weight-falling method.The clonic and tetanic latency of epileptic seizures in mice were measured with behavioral observation.The electroencephalogram(EEG)was analyzed by frontal cortex discharge activity.Brain dry-wet weight method was detected the changes of brain tissue water content.Positive cells of glial fibrillary acidic protein(GFAP)in dentate gyrus(DG)area of hippocampus was counted by immunofluorescence and the expression level of hippocampus transforming growth factor-β2(TGF-β2)protein was detected by western blot.Results As compared with the model group,the low and high-dose of UR could prolong the latency of epileptic seizures(P<0.05),reduce the abnormal discharge of the brain and cerebral edema(P<0.05),decrease the number of GFAP positive cells and the expression of TGF-β2 protein in the hippocampus(P<0.05).Among them,the high-dose group of Uncaria rhynchophylla has a more significant effect compared with the model group.Conclusion The Uncaria rhynchophylla could reduce the susceptibility of early epilepsy after traumatic brain injury,and have a certain degree of neuroprotection,which may be related to the regulation of astrocytes.
