Study on chemical constituents of Tutcheria championi Nakai by UPLC-Q-Exactive

摘要 目的:为了更好地研究与开发民族药石笔木,采用超高效液相色谱-四极杆-静电场轨道阱高分辨质谱联用(UPLC-Q-Exactive)技术对石笔木叶的化学成分进行快速识别和鉴定。方法:利用Agilent C18色谱柱(100 mm×2.1 mm,1.7μm),以乙腈-水为流动相,梯度洗脱,流速0.3mL·min^-1。质谱使用正、负离子模式扫描采集数据,通过精确质量数、二级质谱碎片信息和保留时间,参照文献和对照品对化合物进行鉴定。结果:共筛查出9种化合物,包括7种黄酮类成分和2种有机酸类成分,其中芦丁、金丝桃苷、槲皮素、山奈酚、芹菜素及没食子酸经与对照品比较得到确认,9种化学成分均为该植物中首次报道。结论:该方法可快速分析鉴定石笔木叶中黄酮类和有机酸类成分,为合理开发石笔木叶在医药及保健品方面的应用提供理论依据。 Objective: To establish an ultra high performance liquid chromatography coupled with hybrid quadrupole-orbitrap mass spectrometry ( UPLC-Q-Exactive) method to rapidly identify and authenticate the chemical constituents in the leaves of Tutcheria championi Nakai in order to research and develop this traditional Chinese medicine. Methods: The analysis was performed on Agilent C18 column ( 100mm× 2.1 mm,1.7μm). The mobile phase consisted of acetonitrile and water,and gradient elution was performed at a flow rate of 0. 3 mL·min^-1. The information of the compounds was acquired in positive ion and negative ion mode. The compounds were identified based on their retention time,accurate mass measurements and the mass fragments by comparison with those in the literature and the reference standards. Results: Nine compounds were screened out,including seven flavonoids and two organic acids. Rutin,hyperoside,quercetin,kaempferol,apigenin and gallic acid were unambiguously identified by comparing with the reference standards. All of the nine chemical compounds were reported for the first time in Tutcheria championi Nakai. Conclusion: The method can quickly analyze and identify the flavonoids and organic acids in the leaves of Tutcheria championi Nakai,and provide theoretical basis for the rational development and application of this drug in medicine and health products.
