摘要 针对CMC还原糖法测定饲料中纤维素酶活力存在的问题,本文对浸提时间、待测液酶活力范围、试样空白测定准确性、计算公式等进行了探索。通过在反应结束后添加葡萄糖的方式提高样品本底测定的准确度。该检测方法的定量限为0.8U/g,精密度为3.6%~5.0%。本方法简便、准确、重现性好,可用于饲料中纤维素酶活力的测定。 For the problems that exist in the determination method of cellulase activity in feed,the extraction time,enzyme activity concentration range of diluent,blank determination accuracy and calculation formula were explored in this research.The accuracy of sample background determination was improved by adding glucose after the reaction.The quantitative limit of this detection method was 0.8 U/g,and the precision was 3.6%~ 5.0%.The method was simple,accurate and reproducible,and suitable for the determination of cellulase activity in feed.
