摘要
【目的】探讨五聚素3(PTX3)基因在子宫内膜癌细胞中的表达及其意义。【方法】通过设计合成PTX3特异性的siRNA、对照质粒及空载体,分别转染HEC-1-A细胞,作为PTX3-siRNA组、NC-siRNA组及Blank组,采用RT-PCR 法检测转染后细胞中PTX3 mRNA 的表达,流式细胞术检测细胞周期,MTT法检测细胞增殖活性,划痕实验检测细胞迁移能力,Transwell小室法检测细胞侵袭能力。【结果】PTX3-siRNA组PTX3 mRNA表达水平显著低于NC-siRNA组和Blank组(P<0.05);G1期细胞所占比例显著高于NC-siRNA组和Blank组(P<0.05),S期细胞所占比例、细胞增殖能力、细胞迁移能力、穿膜细胞数显著低于NC-siRNA组和Blank组(P<0.05)。【结论】下调PTX3基因表达可阻碍子宫内膜癌细胞周期进展,抑制细胞增殖、迁移和侵袭能力。
【Objective】To investigate the effects of PTX-3 gene expression on cell cycle regulation,proliferation,migration and invasion of endometrial cancer cells.【Methods】By designing and synthesizing PTX3-specific siRNA,control plasmid and empty vector,HEC-1-A cells were transfected as PTX3-siRNA group,NC-siRNA group and Blank group,respectively.The expression of PTX3 mRNA in transfected cells was detected by RT-PCR.The cell cycle was analyzed by flow cytometry and the cell proliferation activity was measured by MTT assay.The scratch assay was used to reflect cell migration ability,and the Transwell chamber assay was applied to evaluate cell invasion ability.【Results】The expression of PTX3 mRNA in PTX3-siRNA group was significantly lower than that in NC-siRNA group and Blank group (P<0.05).The proportion of cells in G1 phase in PTX3-siRNA group was significantly higher than that in NC-siRNA group and Blank group (P<0.05),while the proportion of S phase cells in PTX3-siRNA group was significantly lower than that of the NC-siRNA group and the Blank group (P<0.05).The cell proliferation ability,cell migration ability and the number of transmembrane cells in PTX3-siRNA group were significantly lower than those in the NC-siRNA group and the Blank group (P<0.05).【Conclusion】 Down-regulation of PTX3 gene expression may impede the progression of endometrial cancer cell cycle and inhibit cell proliferation,migration and invasion.
