肿瘤坏死因子诱导蛋白3基因多态性与变应性鼻炎遗传易感性的关系

Association between Polymorphism of Tumor Necrosis Factor-inducible Protein 3 Gene and Genetic Susceptibility to Allergic Rhinitis

摘要 【目的】探讨肿瘤坏死因子α诱导蛋白3(TNFAIP3)基因多态性与变应性鼻炎(allergic rhinitis,AR)遗传易感性的相关性。【方法】选择2016年1月至2017年8月本院收治的200例AR患者(观察组)和200例健康志愿者(对照组)的血标本作为研究对象。应用限制性片段长度多态性聚合酶链反应(PCR-RFLP)和直接测序的方法对从抗凝血中提取的DNA样本中TNFAIP3基因的4个多态位点(rs5029928,rs9494885,rs610604,rs7753873)进行基因分型。【结果】两组TNFAIP3 基因的4个多态性位点基因分型均符合哈迪温伯格平衡;位点 rs9494885 的等位基因和基因型频率存在统计学差异。观察组的CACA单体型频率明显低于对照组(P<0.05),TCCA单体型频率明显高于对照组(P<0.05)。两组其余指标比较,差异无统计学意义(P>0.05)。【结论】AR与TNFAIP3基因多态性有明显相关性,且 TNFAIP3 的 rs9494885 位点多态性与AR 的易感性密切相关。 【Objective】To investigate the relationship between gene polymorphism of tumor necrosis factorαinduced protein 3 (TNFAIP3) and genetic susceptibility to allergic rhinitis (AR).【Methods】From January 2016 to August 2017,the blood samples of 200 patients with AR (observation group) and 200 healthy volunteers (control group) were selected as the subjects.Four polymorphism loci (rs5029928,rs9494885,rs610604,rs7753873) of TNFAIP3 gene in DNA samples extracted from anticoagulant were genotyped by restriction fragment length polymorphism polymerase chain reaction (PCR-RFLP) and direct sequencing.【Results】The genotypes of four polymorphic loci of TNFAIP3 gene in both groups were in accordance with Hardy Weinberg equilibrium,and there were significant differences in allele and genotype frequencies of rs94885 locus between the two groups.The frequency of ACA haplotype in the observation group was significantly lower than that in the control group (P<0.05),and the frequency of TCCA haplotype was significantly higher than that in the control group (P<0.05).There was no significant difference in other indicators between the two groups (P>0.05).【Conclusion】There is a significant correlation between AR and TNFAIP3 gene polymorphism,and the rs94885 polymorphism of TNFAIP3 is closely related to AR susceptibility.
