摘要
目的研究绿花白千层叶油成分及其抑菌、抗炎活性。方法通过GC-MS对绿花白千层叶油进行分析,峰面积归一化法计算各成分相对百分含量;牛津杯法测定抑菌圈评价其对大肠埃希菌、金黄色葡萄球菌及白色念珠菌的抑菌性;建立脂多糖(LPS)诱导RAW264.7细胞炎症模型,采用酶联免疫吸附法检测细胞上清液中白细胞介素(IL)-1β及肿瘤坏死因子(TNF)-α的表达来考察其抗炎效果。结果本实验鉴定出27个挥发性成分,占峰面积的95.23%,其中桉树醇含量最高,占40.38%;绿花白千层叶油对金黄色葡萄球菌、大肠埃希菌、白色念珠菌的抑菌效果分别属于极敏、高敏、中敏;绿花白千层叶油能显著抑制RAW264.7细胞分泌TNF-α、IL-1β(P<0.01),且具有剂量依赖性。结论绿花白千层叶油成分复杂,且具有较强的体外抑菌、抗炎活性,其抗炎作用可能与阻止RAW264.7细胞向M1型极化,降低TNF-α、IL-1β的表达量有关。
OBJECTIVE To investigate the composition of the leaf oil of Melaieuca viridiflora and its antibacterial and anti-inflammatory activities.METHODS The chemical components of the leaf oil of Melaieuca viridiflora were separated and identified by GC-MS,and the relative content of each component was determined by area nomalization.Through the Oxford cup method, the inhibition of Escherichia coli,Staphylococcus aureus and Candida albicans was evaluated by the inhibition zone.The lipopolysaccharide ( LPS)-induced RAW264.7 cell inflammation model was established.The anti-inflammatory effect of IL-1β and TNF-α was detected by enzyme-linked immunosorbent assay ( ELISA).RESULTS In this experiment,28 volatile components were identified,accounting for 95.23% of the total peak area,of which the highest content of eucalyptol was 40.38%.Staphylococcus aureus was extremely sensitive to the leaf oil of Melaieuca viridiflora,and Escherichia coli was highly sensitive to it.Candida albicans is moderately sensitive to the leaf oil of Melaieuca viridiflora.The leaf oil of Melaieuca viridiflora can significantly inhibit the secretion of TNF-α and IL-1β( P < 0.01 ) by RAW264.7 cells and has a dose dependency.CONCLUSION The leaf oil of Melaieuca viridiflora has a complex composition and strong antibacterial and anti-inflammatory activity,and its anti-inflammatory effect may be related to preventing the differentiation of RAW264.7 cells into M1 type and reduce the expression of TNF-α,IL-1β.
