摘要
目的 探讨前列腺癌组织中转录因子叉头框蛋白P1(FOXP1)和叉头框蛋白Q1(FOXQ1)的表达水平,并分析其与临床病理参数和预后的关系.方法 选取2016年5月至2018年5月于荆州市第三人民医院确诊并接受前列腺癌根治性手术的前列腺癌患者76例其标本为研究对象,以癌组织标本为前列腺癌组,对应癌旁组织标本为癌旁组,免疫组织化学法检测前列腺癌组织及癌旁组织中FOXP1和FOXQ1蛋白阳性表达率;分析FOXP1和FOXQ1在前列腺癌组织中的表达水平及与临床病理参数的关系;采用Spearman相关分析FOXP1和FOXQ1的相关性;对患者进行5年随访,采用Kaplan-M eier分析其与预后的关系,Cox回归分析影响前列腺癌患者生存时间的相关因素.结果 癌组织标本中FOXP1阳性率(67.11%)显著高于癌旁组织阳性率(23.68%),差异有统计学意义(P<0.05);癌组织标本中FOXQ1阳性率(63.16%)显著高于癌旁组织阳性率(26.32%),差异有统计学意义(P<0.05);FOXP1在前列腺癌组织中的表达与年龄、前列腺癌组织学分级(Gleason分级)评分无关,与肿瘤大小、浸润深度、前列腺癌特异性抗原(PSA)水平、淋巴结转移相关(P<0.05);FOXQ1在前列腺癌组织中的表达与年龄、肿瘤大小、浸润程度、PSA水平均无关,与Gleason评分、淋巴结转移相关(P<0.05);相关性分析结果显示,二者在前列腺癌中的表达呈正相关(r=0.618,P<0.05).Kap-lan-M eier分析结果显示,FOXP1、FOXQ1阴性表达者无进展生存期(PFS)、总体生存时间(OS)累积生存率高于阳性者,差异均有统计学意义(P<0.05);Cox回归分析显示FOXP1、FOXQ1阳性表达均是影响前列腺癌患者生存时间的危险因素(P<0.05).结论 前列腺癌组织中转录因子FOXP1和FOXQ1阳性表达率均较高,且与疾病的发生、发展及预后相关.
Objective To investigate the expression levels of transcription factor fork-framed protein P1 (FOXP1) and fork-framed protein Q1 (FOXQ1) in prostate cancer tissues,and to analyze their relationships with clinical pathological parameters and prognosis. Methods From May2016 to May2018,76 patients with prostate cancer who were diagnosed in our hospital and underwent radical prostatectomy were selected.Cancer tissues were taken as the prostate cancer group,and the corresponding paired normal tissues were taken as the normal group.The positive expression rates of FOXP1 and FOXQ1 protein in prostate cancer tissues and adjacent tissues were detected by immunohistochemistry.The expression level of FOXP1 and FOXQ1 in prostate cancer tissues and their relationship with clinical pathological parameters were analyzed.Spearman correlation analysis was used to analyze the correlation between FOXP1 and FOXQ1.The patients were followed up for 5 years,Kaplan-Meier was used to analyze their relationships with prognosis,Cox analysis was used to analyze the related factors affecting the survival time of patients with prostate cancer. Results The positive rate of FOXP1 in cancer tissues (67.11%) was significantly higher than that in normal tissues (23.68%)( P<0.05 ).The positive rate of FOXQ1 in cancer tissues (63.16%) was significantly higher than that in normal tissues (26.32%)( P<0.05 ).The expression of FOXP1 in prostate cancer tissues was not related with age and Gleason score,but correlated with tumor size,depth of invasion,PSA level and lymph node metastasis ( P<0.05).The expression of FOXQ1 in prostate cancer tissues was not related with age,tumor size,invasion degree and PSA level,but was related to Gleason score and lymph node metastasis ( P<0.05).Correlation analysis showed that there was a positive correlation between the two expressions in prostate cancer ( r = 0.618, P<0.05 ).Kaplan-Meier analysis showed that PFS and OS in FOXP1 negative expression patients were higher than PFS and OS in positive expression,PFS and OS in FOXP1 negative expression patients were higher than PFS and OS in FOXP1 positive expression patients ( P<0.05).Cox regression analysis showed that FOXP1 and FOXQ1 were both risk factors affecting the survival time of prostate cancer patients. Conclusion The positive expression rates of transcription factors FOXP1 and FOXQ1 in prostate cancer tissues are higher,and they are related to the occurrence,development and prognosis of the disease.
