摘要
目的探索地衣芽胞杆菌BL63516 对DSS 诱导的小鼠结肠炎的缓解作用和可能机制,为临床疾病的治疗提供新思路。方法用BALB/C 雌性小鼠构建动物模型,将30 只小鼠分为CON 组、DSS 组和BL 组,每组10 只,除CON 组外,其余两组均给予3%( m/v) DSS 自由饮用,并给予地衣芽胞杆菌BL63516 干预。第8 天摘眼球处死小鼠,取血、结肠、粪便样本。以ELISA 法检测小鼠血清和结肠研磨液中的细胞因子。用PCR-变性梯度凝胶电泳( PCR-DGGE)方法分析小鼠肠道菌群结构。结果DSS 组小鼠血清IL-10 及结肠MPO 水平均与CON 组有明显差异,地衣芽胞杆菌干预后有不同程度改变;使用非加权成对算术平均算法( UPGMA)对PCR-DGGE 图谱中各泳道条带类型聚类分析结果显示各组小鼠结肠菌群结构具有显著差异,其中BL 组增加双歧杆菌属Bifidobacterium saguini 及疣微菌门的噬黏蛋白阿克曼菌( Akkermansiamuciniphila)数量。结论地衣芽胞杆菌可以有效改善DSS 诱导的结肠炎症状,其作用机制与肠屏障功能和肠道菌群的调节有密切关系。
Objective To explore the effect of Bacillus licheniformis BL63516 on DSS-induced colitis in mice and provide new ideas for clinical treatment of the disease. Methods Thirty mice were divided into group CON, group DSS or group BL. The groups DSS and BL were given 3% DSS(m/v) to drink freely, while group BL was additionally fed Bacillus licheniformis. The mice were killed at the 8th day to collect samples of blood, colon and feces. The cytokine levels in the serum and colonic tissue were detected with ELISA. Denaturing gradient gel electrophoresis(DGGE) was used to analyze the intestinal microflora of mice. Results The levels of serum IL-10 and colonic MPO in group DSS were significantly different from those in the control group, which were improved after the intervention of Bacillus licheniformis BL63516. The results of cluster analysis on the types of bands in each lane in PCR-DGGE atlas using non-weighted arithmetic averaging algorithm(UPGMA) showed that there were different intestinal floras in different groups. Group BL showed the bands of Bifidobacterium saguini and Akkermansia muciniphila. Conclusion Bacillus licheniformis BL63516 can effectively improve the symptoms of colitis induced by DSS;its mechanism is closely related to intestinal barrier function and regulation of gut microbiota.
