五聚素3在子宫内膜癌组织中的表达及与子宫内膜癌恶性程度和预后的关系

Expression of PTX3 in endometrial carcinoma and its relationship with malignancy and prognosis of endometrial cancer

目的探讨五聚素3(PTX3)在子宫内膜癌组织中的表达及其与子宫内膜癌恶性程度和预后的关系。方法将2015年1月至2016年12月中国石油集团公司中心医院收治的167例子宫内膜癌患者作为研究对象,收集患者子宫内膜癌组织(1.5cm×1.0cm×1.0cm)作为检测样本,另取癌旁组织(距癌组织边缘≥5cm)作为对照组织,分别采用RT-PCR及westernblot检测子宫内膜癌组织中PTX3mRNA及蛋白的表达水平,并分析PTX3表达水平与子宫内膜癌恶性程度和预后的关系。结果子宫内膜癌组患者组织中PTX3mRNA表达水平(4.93±0.89)明显高于相应癌旁组织(0.96±0.31),差异具有统计学意义(P<0.05),且PTX3蛋白表达水平(1.98±0.54)也明显高于相应癌旁组织(0.43±0.26),差异也具有统计学意义(P<0.05)。PTX3表达水平与子宫内膜癌患者病理分期、分化程度、淋巴结转移、浸润肌层深度有关(P<0.05),而与年龄、病理类型无关(P>0.05)。PTX3mRNA高表达组(≥4.93)与低表达组(<4.93)的无进展生存期分别为10个月(95%CI为7.8~12.1个月)和19个月(95%CI为13.9~23.8个月),二者相比差异有统计学意义(P<0.05);总生存期分别为14个月(95%CI为10.3~17.0个月)和24个月(95%CI为17.6~29.2个月),二者相比差异也有统计学意义(P<0.05)。结论PTX3表达水平升高与子宫内膜癌的恶性程度有关,可能对子宫内膜癌的预后判断具有一定的指导价值。

Objective To investigate the expression of PTX3 in endometrial carcinoma and its relationship with the degree of malignancy and prognosis of endometrial cancer.Methods From January2015 to December2016,167 patients with endometrial cancer admitted to our hospital were included in the study.The endometrial cancer tissues of patients (1.5 cm ×1.0 cm ×1.0 cm) were collected as test samples.And the adjacent tissues (≥5 cm from the edge of the cancer tissue) were taken as control tissues.The expression levels of PTX3 mRNA and protein in endometrial carcinoma tissues were detected by RT-PCR and western blot,respectively.The relationship between the expression level of PTX3 and the degree of malignancy and prognosis of endometrial cancer was analyzed.Results The expression of PTX3 mRNA in the tissues of patients with endometrial cancer was significantly higher than that of the adjacent tissues (4.93±0.89 vs 0.96±0.31,P < 0.05).The expression level of PTX3 protein was significantly higher than that of the corresponding adjacent tissues (1.98±0.5 vs 0.43±0.26,P < 0.05).The expression level of PTX3 was correlated with the pathological stage,differentiation degree,lymph node metastasis and depth of invasion of endometrial cancer patients ( P < 0.05) with no relation to the age and pathological type (P>0.05).The progression-free survival of the PTX3 mRNA high expression group (≥4.93) and the low expression group (<4.93) were10 months (95% CI 7.8- 12.1 months) and19 months (95% CI13.9-23.8) respectively.The difference between the two was statistically significant ( P < 0.05).The overall survival was14 months (95% CI10.3-17.0 months) and24 months (95% CI17.6-29.2) respectively.The difference between the two was statistically significant ( P < 0.05).Conclusions The elevated expression level of PTX3 is related to the malignant degree of endometrial cancer,which may have certain guiding value for the prognosis of endometrial cancer.
