支气管哮喘患儿外周血SDF-1、CXCR4水平与气道炎症和气流受限的关系 被引量:12

Relationship between SDF-1 and CXCR4 levels in peripheral blood with airway inflammation and airflow limitation in children with bronchial asthma

摘要 目的检测支气管哮喘患儿外周血基质细胞衍生因子-1(SDF-1)、CXC趋化因子受体4(CXCR4)水平,研究其与气道炎症和气流受限的关系。方法选取2016年12月-2017年12月本院确诊收治的支气管哮喘患儿78例,选取同期健康体检者60例作为对照组,采用酶联免疫吸附法(ELISA)法检测血清SDF-1水平,采用流式细胞术检测外周血CXCR4水平,利用肺功能仪检测2组受试者1秒用力呼气量(FEV1)、用力肺活量(FVC),检测2组受试者诱导痰液中炎性细胞数目。结果研究组FEV1占预计值百分比、FEV1/FVC较对照组显著降低(P<0.05);研究组患儿诱导痰中细胞总数、中性粒细胞、嗜酸粒细胞、淋巴细胞数目显著高于正常儿童(P<0.05),研究组患儿巨噬细胞数目显著低于正常儿童,研究组外周血SDF-1、CXCR4水平显著高于对照组(P<0.05);支气管哮喘患儿外周血SDF-1、CXCR4水平与FEV1占预计值百分比、FEV1/FVC、巨噬细胞数目负相关(P<0.05);与细胞总数、中性粒细胞数目、嗜酸粒细胞数目、淋巴细胞数目正相关(P<0.05)。结论支气管哮喘患儿外周血SDF-1、CXCR4水平显著高于对照组,与支气管哮喘患儿气道炎症及气流受限关系密切。 Objective To detect the levels of stromal cell derived factor-1(SDF-1)and CXC chemokine receptor-4(CXCR4)in peripheral blood of children with bronchial asthma,and to study their relationships with airway inflammation and airflow restriction.Methods 78 cases of bronchial asthma admitted to our hospital from December 2016 to December 2017 were selected,and 60 healthy people during the same period were selected as the control group.Enzyme-linked immunosorbent assay(ELISA)was used to detect serum SDF-1 level,the level of CXCR4 in peripheral blood was detected by flow cytometry,the forced expiratory volume in one second(FEV 1)and forced vital capacity(FVC)of the two groups were measured by the pulmonary function instrument,and the number of inflammatory cells in induced sputum of the two groups was detected.Results The percentage of FEV 1 in estimated value and the FEV 1/FVC in the study group were significantly lower than those in the control group(P<0.05).The total number of cells,neutrophils,eosinophils and lymphocytes in the study group were significantly higher than those in the control group(P<0.05),the number of macrophages in the study group was significantly lower than that in the control group,and the levels of SDF-1 and CXCR4 in the peripheral blood of the study group were significantly higher than those of the control group(P<0.05).The levels of SDF-1 and CXCR4 in peripheral blood of children with bronchial asthma were negatively correlated with the percentage of FEV 1 in estimated value,FEV 1/FVC and the number of macrophages(P<0.05),and it was positively correlated with the total number of cells,the numbers of neutrophils,eosinophils and lymphocytes(P<0.05).Conclusion The levels of SDF-1 and CXCR4 in peripheral blood of asthmatic children are significantly higher than those in the study group,and the levels of SDF-1 and CXCR4 in peripheral blood are closely related to airway inflammation and airflow limitation.
