摘要
目的探讨Frat1能否通过Wnt/β-catenin信号通路对非小细胞肺癌A549细胞的增殖、凋亡产生影响。方法RT-PCR检测非小细胞肺癌组织及正常肺组织中Frat1、β-catenin的表达;CCK-8试验检测Frat1沉默组和Frat1正常表达组非小细胞肺癌A549细胞的增殖;TUNEL免疫荧光法检测Frat1沉默组和Frat1正常表达组非小细胞肺癌A549细胞的凋亡;Western Blot检测Frat1沉默组和Frat1正常表达组非小细胞肺癌A549细胞β-catenin蛋白的表达。结果非小细胞肺癌组织中Frat1、β-catenin基因的表达显著高于正常肺组织(P<0.05);Frat1沉默组细胞的增殖能力显著弱于Frat1正常表达组(P<0.05);Frat1沉默组细胞的凋亡显著高于Frat1正常表达组(P<0.05);Frat1正常表达组细胞β-catenin蛋白的表达显著高于Frat1沉默组(P<0.05)。结论Frat1与β-catenin在非小细胞肺癌的表达被激活;Frat1能促进非小细胞肺癌A549细胞的增殖,抑制其凋亡,这种作用可能是通过激活Wnt/β-catenin信号通路而实现的。
Objective To investigate whether Frat1 can affect the proliferation and apoptosis of non-small cell lung cancer A549 cells through Wnt/β-catenin signaling pathway.Methods RT-PCR was used to detect the expression of Frat1 andβ-catenin in non-small cell lung tissues and normal lung tissues.CCK-8 assay was used to detect the proliferation of non-small cell lung cancer A549 cells with Frat1 silencing and normal expression of Frat1.TUNEL immunofluorescence assay was used to detect the apoptosis of non-small cell lung cancer A549 cells with Frat1 silencing and normal expression of Frat1.Western Blot was used to detectβ-catenin protein expression in non-small cell lung cancer A549 cells with Frat1 silencing and normal expression of Frat1.Results The expression of Frat1 andβ-catenin genes in non-small cell lung tissues was significantly higher than that in normal lung tissues(P<0.05).The proliferation of Frat1 silenced cells was significantly weaker than that of normal cells(P<0.05).The apoptosis of Frat1 silenced cells was significantly higher than that of normal cells(P<0.05).The expression ofβ-catenin protein in normal cells of Frat1 was significantly higher than that of Frat1 silenced cells(P<0.05).Conclusion The expression of Frat1 andβ-catenin in non-small cell lung cancer is activated.Frat1 can promote the proliferation of non-small cell lung cancer A549 cells and inhibit its apoptosis,which may be achieved by activating Wnt/β-catenin signaling pathway.
