N-乙酰半胱氨酸对AGEs诱导的HK-2细胞IL-1β、IL-6的抑制作用

摘要 目的:探讨N-乙酰半胱氨酸(Nac)对终末糖基化产物(AGEs)诱导的人肾小管上皮细胞(HK-2细胞)IL-1β(白介素-1β)、IL-6(白介素-6)的抑制作用。方法:将体外培养的HK-2细胞分成正常对照组(NG组)、AGEs组、药物干预组1(N1组)、药物干预组2(N2组),培养72 h,应用反转录实时定量聚合酶链式反应(RT-qP CR)方法检测IL-1β和IL-6 mRNA表达,酶联免疫吸附法(ELISA)测定培养上清中细胞因子的蛋白浓度。蛋白质印迹法(Western blot)检测NF-κB/p65的含量。结果:NG组、AGEs组、N1组和N2组细胞内IL-1β和IL-6 mRNA和蛋白表达,以及NF-κB/p65含量差异均有统计学意义;与NG组比较,AGEs组细胞内IL-1β和IL-6mRNA和蛋白表达明显增加。N1、N2组较AGEs组表达水平下降,AGEs组NF-κB/p65含量高于其他组,差异均具有统计学意义(均P <0. 05)。结论:AGEs可诱导HK-2细胞表达IL-1β和IL-6,NF-κB信号通路可能参与了此过程。
