摘要
目的:探索羟基积雪草苷对人宫颈癌细胞C-33A/CaSki细胞凋亡影响。方法:不同浓度的羟基积雪草苷干预C-33A/CaSki细胞,MTT法检测药物对细胞的增殖抑制作用。TUNEL技术检测细胞的凋亡率。ELISA法测定细胞中Bcl-2、Bclxl及Bax的表达变化。QRT-PCR检测Bcl-2、Bcl-xl及Bax的mRNA表达改变。结果:Madecassoside对C-33A/CaSki细胞具有增殖抑制作用,并呈现浓度时间依赖性。不同浓度的Madecassosid能引起细胞凋亡率增加。ELISA与QRT-PCR结果显示,Madecassoside能下调抑制凋亡的因子Bcl-2、Bcl-xl的表达,上调能促进凋亡的因子Bax的表达,在mRNA上呈现相同变化。结论:Madecassoside可以通过上调促进凋亡的因子的表达,下调对凋亡具有抑制作用的因子表达从而发挥抗肿瘤作用。
Objective:To explore the effect of hydroxycampanulin on apoptosis of C-33A/CaSki cells in human cervical cancer cells.Methods:Different concentrations of hydroxycumulus glycoside interfered with C-33A/CaSki cells,and MTT assay was used to detect the inhibitory effect of drugs on cell proliferation.TUNEL assay was used to detect the apoptosis rate of cells.The expression of Bcl-2,Bcl-xl and Bax in cells was determined by ELISA.MRNA expression changes of Bcl-2,Bcl-xl and Bax were detected by QRT-PCR.Results:Madecassoside had an inhibitory effect on proliferation of C-33A/CaSki cells in a concentration time dependent manner.Different concentrations of Madecassosid could increase apoptosis rate.ELISA and QRT-PCR results showed that Madecassoside could downregulate the expression of Bcl-2 and Bcl-xl,which inhibited apoptosis,and up-regulated the expression of Bax,which promoted apoptosis,showing the same change in mRNA.Conclusion:Madecassoside could play an anti-tumor role by up-regulating the expression of apoptotic factors and down-regulating the expression of apoptotic inhibitory factors.
