摘要
运用荧光光谱、同步荧光光谱、紫外可见吸收光谱结合分子模拟技术,研究漆黄素与胰蛋白酶的相互作用及抗氧化活性。结果表明,漆黄素能够与胰蛋白酶作用形成复合物并导致胰蛋白酶内源性荧光发生静态猝灭,该结合主要由疏水作用力驱动,25℃时,结合常数为7.94×10^4L·mol^-1,二者存在一个结合位点。漆黄素对DPPH自由基和超氧阴离子自由基都有较强的清除能力,而胰蛋白酶能够在一定程度上抑制漆黄素对DPPH自由基和超氧阴离子自由基的清除作用,从而影响漆黄素的抗氧化活性。
The interaction of fisetin with trypsin and the antioxidant activity of fisetin were studied by using fluorescence spectra,synchronous fluorescence spectra,UV visible absorption spectra and molecular simulation techniques.The results showed that fisetin could bind with trypsin to form a compound and lead to static quenching of endogenous fluorescence of trypsin.The binding was mainly driven by hydrophobic forces with a binding constant of 7.94×10^4 L·mol^-1 at 298 K,and there was one binding site between them.Fisetin exhibited a strong scavenging ability on DPPH free radical and superoxide anion radical.Trypsin in a certain extent inhibited the scavenging effect of fisetin on DPPH free radical and superoxide anion free radical,and affected the antioxidant activity of fisetin.
