燧心胶囊对心肌梗死后心衰大鼠心功能及心肌肌浆网钙泵基因表达的影响

Effect of Suixin Capsule on Cardiac Function and Gene Expression of Calcium Pump in Myocardial Sarcoplasmic Reticulum in Rats with Heart Failure after Myocardial Infarction

目的观察燧心胶囊对心肌梗死后心衰大鼠心功能及心肌肌浆网钙泵(SERCA)、受磷蛋白(PLB)基因表达的影响。方法选取80只大鼠用左前降支冠状动脉结扎法制备大鼠心衰模型,将造模成功大鼠分为模型对照组,卡托普利组,燧心胶囊低、中、高剂量组,同时设置没有进行冠状动脉结扎大鼠10只为假手术组。燧心胶囊低、中、高剂量组依次灌胃2.7、5.4和10.8g/kg的燧心胶囊药液,卡托普利组给予5.83mg/kg卡托普利,每日1次,连续8周。假手术组和模型对照组给予等量生理盐水。实验结束后检测大鼠心功能和血流动力学,同时检测心肌中SERCA和PLB mRNA的表达。结果与假手术组比较,模型组大鼠心功能和血流动力学参数差异有统计学意义(P<0.05),表明模型制备成功。与模型组比较,卡托普利组和各燧心胶囊治疗组大鼠LVESD、LVEDD、LVSP和LVEDP降低,LVEF、LVFS、+dp/dtmax和-dp/dtmax增高,大鼠心肌组织中SERCA mRNA表达升高,PLB mRNA表达降低,各燧心胶囊治疗组指标水平随药物剂量增加而变化,差异具有统计学意义(P<0.05)。结论燧心胶囊可以增强心衰大鼠射血能力,改善心功能,其机制可能与上调心肌细胞中SERCA基因的表达有关。

Objective:To investigate the effect of Suixin Capsule on cardiac function and gene expression of calcium pump in myocardial sarcoplasmic reticulum in rats with heart failure after myocardial infarction.Methods: 80 rats were selected to establish heart failure model by ligation of left anterior descending coronary artery.The successful rats were divided into the model control group,captopril group,low,medium and high dose groups of Suixin Capsule and sham operation group without ligation of coronary artery.The low,middle and high dose groups were given 2.7,5.4,10.8 g/kg respectively by intragastric administration.The captopril group was given 5.83 mg/kg captopril for 8 weeks.Sham operation group and the model control group were given the same amount of saline.At the end of the experiment,cardiac function and hemodynamics were measured,and the expressions of SERCA and PLB in myocardium were detected.Results:Compared with sham-operated group,cardiac function and hemodynamic parameters in the model group had significant difference(P < 0.05),which indicating that the model was successfully prepared.Compared with the model group,LVESD,LVEDD,LVSP and LVEDP in captopril group and Suixin Capsule treatment groups decreased;LVEF,LVFS,+dp/dtmax and-dp/dtmax increased,and SERCA expression in rat myocardium increased.The level of indicators in the treatment groups varied with the increase of drug dosage,and the difference was statistically significant(P < 0.05).Conclusion:Suixin Capsule can enhance ejection ability and improve cardiac function in rats with heart failure.Its mechanism may be related to up-regulation of SERCA gene expression in myocardial cells.
