摘要
目的研究针刺对膝骨性关节炎(KOA)大鼠滑膜中神经生长因子(NGF)和P物质(SP)表达的影响,以探讨针刺治疗KOA的镇痛作用机制。方法选取成年SD大鼠75只,采用随机数字表法将大鼠分为5组,各15只,依次为正常组、针刺穴位组、造模组、西乐葆组和针刺非穴组。造模组、西乐葆组、针刺穴位组和针刺非穴位组均采用木瓜蛋白酶注入左膝关节腔的方法制备大鼠KOA模型,治疗1周后,测定各组大鼠滑膜NGF、SP含量。结果造模组NGF、SP含量高于正常组,差异有统计学意义(P<0.01);NGF、SP含量西乐葆组、针刺穴位组和针刺非穴组均显著低于造模组(P<0.05或P<0.01),NGF、SP含量针刺穴位组显著低于针刺非穴组(P<0.05);但西乐葆组与针刺穴位组水平相当,比较无统计学差异(P>0.05)。结论针刺能有效抑制NGF、SP的表达,从而达到镇痛效果。
Objective:To study the effect of acupuncture on the expression of NGF and SP in KOA rats,and to explore its analgesic mechanism.Methods:75 SD rats were randomly divided into the normal group,the model group,Celebrex group,acupoint group and non-acupoint group,15 rats in each.KOA model rats were made by injecting papain into the left knee joint cavity of rats.After one week of treatment,the content of NGF and SP in synovium was calculated.Results:The content of NGF and SP in the model group was higher than that in the normal group(P < 0.01);those in acupoint group,Celebrex group and non-acupoint group were significantly lower than the model group(P < 0.05 or P < 0.01);that in acupoint group was significantly lower than that in the non-acupoint group.However,there was no significant difference between Celebrex group and acupoint group(P > 0.05).Conclusion:Acupuncture can effectively inhibit the expression of NGF and SP,thus achieving analgesic effect.
