摘要 探讨辣木叶提取物对胰脂肪酶的抑制作用,为开发辣木叶天然胰脂肪酶抑制剂提供理论依据。在单因素试验的基础上,用正交试验优化提取条件,研究辣木叶提取物对胰脂肪酶活性的抑制作用,并结合Dixon图和Lineweaver-Burk图探讨其抑制类型。结果表明最佳提取条件为:料液比1∶50(g/mL)、提取液乙醇浓度70%、超声功率400W、超声处理时间600s、提取温度50℃;该条件下辣木叶粗提物对胰脂肪酶活力的抑制率为79.15%;辣木叶提取物对胰脂肪酶的抑制作用为非竞争性抑制。辣木叶提取物对胰脂肪酶的活性有明显抑制作用。 In order to provide basis for employing Moringa oleifera as lipase inhibitor, the inhibitory effect of extracts of Moringa oleifera leaves on lipase activity was investigated. Based on single-factor experiments, orthogonal experiments were explored to obtain optimum conditions. The effect of the extract of Moringa oleifera on the inhibition of pancreatic lipase activity and the inhibition type Dixon and Lineweaver-Burk graphs were explored. The optimum extracting conditions were established as following:solid to liquid ratio was 1 ∶ 50(g/mL), ultrasonic power was 400 W, for 600 s with 70 % ethanol, at 50 ℃. Under these conditions,the inhibitory rate of extracts of moringa leaves on pancreatic lipase was 79.15 %. The inhibition type was noncompetitive. The extract of Moringa oleifera leaves had inhibitory effect on pancreatic lipase activity.
