基于UPLC/Q-TOF MS技术对延胡索醋法炮制前后化学成分的研究 被引量:11

摘要 建立了超高效液相色谱-串联四极杆飞行时间质谱(ACQUITY UPLCTM I-Class Xevo G2-XS Q-TOF MS)法快速鉴定醋法炮制前后延胡索化学成分差异的影响,使用UNIFI软件对数据进行了分析,以多元统计分析VIP>1,P<0.05作为判断标准,共发现28个延胡索生物碱标记物,除黄连碱、隐品碱醋制后含量降低以外,其余生物碱类成分均有所升高,尤其是延胡索甲素、四氢黄连碱、小檗碱、二氢血根碱、去氢海罂粟碱等。结果表明,应用UPLC/Q-TOF MS技术能够快速检定炮制后延胡索药材的化学成分差异,通过比较化学成分差异为临床疗效研究提供重要的参考。 A UPLC/Q-TOF MS(ACQUITY UPLCTM I-Class Xevo G2-XS Q-TOF MS)method was established in this study to rapidly identify the chemical components from Rhizoma Corydalis before and after vinegar processing.The data was analyzed by UNIFI software with multivariate statistical analysis of VIP>1,P<0.05 as the judgment standard.Total of 28 corydalis alkaloids were identified as marker compounds.The content of Coptisine and Cryptopine were lower after vinegar processing while the content of the rest alkaloids were higher after vinegar processing,especially Corydaline,Tetrahydrocoptisine,Berberine,Dihydrosanguinarine,Tetradehydroglaucine.The results showed that the application of UPLC/Q-Tof MS technology could rapidly identify the chemical components of processed rhizoma corydalis and the established method was effective for the analysis of the components of Traditional Chinese Medicine.
