叶下珠提取物对DPPH和亚硝酸盐的清除作用研究 被引量:2

Scavenging Effect of Extracts from Herba Phyllanthi Urinariae on DPPH and Nitrite

摘要 【目的】考察叶下珠提取物的制备工艺及其对2,2-二苯基-1-苦肼基(DPPH)自由基和亚硝酸盐的清除作用。【方法】采用乙醇回流法和液液萃取法获得叶下珠提取物,通过Folin-Ciocaileu比色法测定总多酚的含量,DPPH法测定叶下珠提取物对DPPH自由基的清除率,N-(1-萘基)乙二胺分光光度法测定叶下珠提取物清除亚硝酸盐的能力。【结果】提取得到叶下珠提取物浸膏约40.10g,测得浸膏中总多酚占51.60%。叶下珠提取物清除DPPH自由基的IC50为0.089mg/mL,清除亚硝酸盐的IC50为0.093mg/mL。【结论】采用液液萃取的方法可有效得到叶下珠提取物,操作简单方便,总多酚得率较高。叶下珠提取物对DPPH自由基和亚硝酸盐有较强的清除作用。 Objective To investigate the preparation process of extracts from Herba Phyllanthi Urinariae,and their scavenging effect on 2,2-dipheny1-1-picrylhydrazy1(DPPH)free radical and nitrite.Methods Ethanol reflux method and liquid-liquid extraction method were used to obtain the extracts from Herba Phyllanthi Urinariae.The content of total polyphenols was determined by Folin-ciocaileu colorimetry.The scavenging rate for DPPH free radical of Herba Phyllanthi Urinariae extracts was determined by DPPH method.The nitrite scavenging activity of Herba Phyllanthi Urinariae was determined by N-(1-naphthyl)ethylenediamine spectrophotometry.Results Herba Phyllanthi Urinariae extracts extractum achieved was about 40.10 g,the total polyphenols in the extractum accounting for 51.60%.The IC50 for Herba Phyllanthi Urinariae extracts scavenging DPPH radical and nitrite was 0.089 and 0.093 mg/mL,respectively.Conclusion The liquid-liquid extraction method is effective for obtaining Herba Phyllanthi Urinariae extracts,which is simple and convenient to operate and has higher yield.And Herba Phyllanthi Urinariae extracts have strong scavenging effect on DPPH free radical and nitrite.
