桃实蝇的DNA条形码鉴定

摘要 桃实蝇(Bactrocera zonata)严重危害桃、番石榴、番荔枝等经济作物,被我国列为进境植物检疫性有害生物。为明确桃实蝇各虫态的重要形态鉴定特征,并将DNA条形码技术应用于桃实蝇的鉴定,本研究针对采自于印度旁遮普邦的桃实蝇卵、幼虫、蛹和成虫样品进行了形态观察和分子鉴定。结果表明:桃实蝇成虫整体呈红褐色,头部具一对卵圆形黑色颜面斑。翅除前缘带和臀条外几乎透明,r2+3室端部具“痣状”小褐斑,延伸至r4+5室端部。实蝇成虫及蛹样品的COI条形码序列与GenBank中收录的桃实蝇序列相似度在99.24%~100%之间;基于COI条形码构建的系统发育树显示本研究所有样品和桃实蝇聚在一个分支上。本研究表明DNA条形码技术可有效应用于桃实蝇的准确鉴定。 Bactrocera zonata (Tephritidae:Bactrocera) severely damages economic crops,including peach, guava and cherimoya etc.So,it has been listed as entry plant quarantine pest in China.To acquire each stage key morphological characteristic of B.zonata and apply DNA barcoding on the molecular identification on B.zonata,the eggs,larvae,pupae and adults of B.zonata from Punjab,India were observed and identified by DNA barcode.The results showed that B.zonata is predominantly red-brown in color and has a pair of oval,black facial spots.The wing is almost hyaline except the costal band and anal vein with a small,isolated brown apical spotat at apex of cell r2+3 and extending to cell r4+5.The COI barcodes sequences similarity between adults,pupae and relevant B.zonata in GenBank were 99.24%-100%.The phylogenetic tree constructed by COI barcode showed that all the samples in this study formed into a clade with B.zonata in GenBank.This study suggested that DNA barcoding could be as an accurate diagnostic method for B.zonata.
