摘要
目的探讨血压级别不同的新疆哈萨克族原发性高血压患者T淋巴细胞上电压依赖性钾通道(Kv1.3钾通道)表达的差异。方法从2018年1-8月,在新疆医科大学第一附属医院、乌鲁木齐市友谊医院和乌鲁木齐市达坂城区柴窝堡社区卫生服务中心选取初诊且未经治疗的哈萨克族高血压患者90例,其中高血压1级组30例,年龄(49.2±9.1)岁,高血压2级组30例,年龄(53.2±8.3)岁,高血压3级组30例,年龄(53.1±9.9)岁;血压正常的哈萨克族健康体检者30名(男女各15名)为对照组,年龄(49.0±9.8)岁。收集静脉血,分离T淋巴细胞,采用逆转录聚合酶链反应(RT-PCR)技术和Western blot技术检测T淋巴细胞Kv1.3钾通道的基因和蛋白表达量及T淋巴细胞肿瘤坏死因子α(TNF-α),白细胞介素6(IL-6)基因表达量。结果与对照组相比,高血压1、2、3级组的Kv1.3钾通道mRNA相对表达量逐渐增高[(1.55±0.54)、(1.75±0.72)、(3.12±1.40)比(1.08±0.49),P<0.05];与对照组相比,高血压1、2、3组的Kv1.3钾通道蛋白相对表达量逐渐增高[(0.17±0.04)、(0.18±0.06)、(0.40±0.17)比(0.11±0.04),P<0.05];与高血压1、2级组相比,高血压3级组Kv1.3钾通道mRNA及蛋白相对表达明显增加(P<0.05)。与对照组(0.42±0.21)相比,高血压2级组(1.82±0.71)、高血压3级组(2.33±1.02)TNF-αmRNA相对表达增高(P<0.05);与对照组(0.27±0.11)相比,高血压1级组(1.10±0.64)、高血压2级组(2.68±1.26)、高血压3级组(2.99±1.57)IL-6 mRNA相对表达增高(P<0.05)。结论 T淋巴细胞上的Kv1.3钾通道在新疆哈萨克族不同级别高血压患者中的表达都增加,其中在高血压3级患者当中的表达增加更为明显。
Objective To investigate the different expression of Kv1.3 potassium channel in T lymphocytes of hypertensives with different grades in Kazak of Xinjiang. Methods From January to August 2018, 90 Kazak hypertensives who were initially diagnosed and untreated in First Affiliated Hospital of Xinjiang Medical University, Friendship Hospital of Urumqi and Community Health Service Center of Chaiwobao in Dabancheng District of Urumqi were recuited. The patients were divided to grade 1 [aged(49.2±9.1) years], grade 2 [aged(53.2±8.3) years], and grade 3 [aged(53.1±9.9) years] group, with 30 cases in each group. The control group consisted of 30 healthy Kazak subjects with normal blood pressure, aged(49.0±9.8) years. There were 15 males and 15 females in each group. Venous blood was collected and T cells were isolated. Gene expressions of Kv1.3 potassium ion channel, tumor necrosis factor-α(TNF-α) and interleukin 6(IL-6) in T cells were detected by real-time quantitative polymerase chain reaction(PCR) and the protein expression of Kv1.3 potassium ion channel by Western blot respectively. Results Compared with the control group(1.08±0.49, 0.11±0.04), the relative mRNA and protein expression of Kv1.3 potassium channel in grade 1 group(1.55±0.54, 0.17±0.04), grade 2 group(1.75±0.72, 0.18±0.06) and grade 3 group(3.12±1.40, 0.40±0.17) increased gradually(all P<0.05). As compared with hypertension grade 1 and grade 2 group, the relative mRNA and protein expression of Kv1.3 potassium channel in grade 3 group increased significantly(P<0.05). The levels of TNF-α mRNA in the grade 2 group(1.82±0.71) and the grade 3 group(2.33±1.02) were higher than those in the control group(0.42±0.21). In addition, the relative expression of IL-6 mRNA in the grade 1 group(1.10±0.64), grade 2 group(2.68±1.26) and grade 3 group(2.99±1.57) was higher than that in the control group(0.27±0.11, P<0.05). Conclusion In Kazak of Xinjiang, the expression of Kv1.3 potassium channel in T lymphocytes increased among hypertensives with different grades, especially in grade 3 hypertensives.
