摘要 目的探讨人子宫内膜间充质干细胞(MenSC)移植对局灶节段性肾小球硬化(FSGS)大鼠肾组织肿瘤坏死因子-α(TNF-α)和白细胞介素-6(IL-6)表达的影响.方法SD大鼠随机分成3组:正常组为正常SD大鼠;模型组和移植组分别于单侧肾切除后第7天和第28天尾静脉注射5mg/kg和2mg/kg阿霉素;移植组大鼠于术后第28天尾静脉注射1mlMenSC细胞悬液(1×10^7/mL),而模型组尾静脉注射等剂量生理盐水.生化检测尿蛋白及肾功能变化,同时RT-PCR结合免疫组化检测肾组织TNF-α和IL-6的变化.结果与正常比较,模型组大鼠24h尿蛋白、血肌酐和尿素氮等均显著增高(P<0.01);肾组织炎症因子TNF-α和IL-6的表达水平增高(P<0.01).MenSC移植后FSGS大鼠肾功能各项指标有所改善,肾组织炎症因子TNF-α和IL-6的表达水平也明显降低(P<0.01或P<0.05).结论MenSC移植可改善肾功能,抑制肾组织中炎症因子TNF-α和IL-6的表达,减轻肾脏的炎症损伤. Objective To investigate the effect of human endometrium stem cell(MenSC)transplantation on the expression of renal TNF-α and IL-6 in rats with focal segmental glomerulosclerosis(FSGS). Methods Sprague Dawley rats were randomly assigned to three groups:No intervention was applied to the rats of normal control group. Model group and transplant group were injected with adriamycin 5 mg/kg(on day 7) and 2mg/kg(on day 28)via the tail vein. In addition,the transplant group rats were injected with 1ml MenSC suspension(1×10^7/mL)via the tail vein on day 28.The model group rats were injected with the same dosage of saline in the same way. The urine protein and changes of renal function were evaluated by biochemical detection,and the expression of renal TNF-α and IL-6 were analyzed through RT-PCR and immunohistochemistry. Results Compared with the control group,the 24-hour urinary protein,serum creatinine and urea nitrogen increased remarkably(P<0.01)and the expression levels of renal TNF-α and IL-6 also increased(P<0.01). However,transplantation of MenSC alleviated the various indexes of renal function. Besides,MenSC decreased the expression levels of renal TNF-α and IL-6(P<0.01 or P<0.05). Conclusions MenSC can improve the renal function,and inhibit the expression of inflammatory factors TNF-α and IL-6 in renal tissue,improve renal inflammation.
