摘要
目的研究梅毒螺旋体对新西兰兔睾丸的病理损伤及其对精子的影响。方法用梅毒螺旋体(Nichols株)及其灭活株分别接种3个月左右的新西兰兔睾丸各8只和4只,接种后定期观察新西兰兔睾丸的形态学改变并获取不同时间点的兔睾丸组织观察其病理变化及精子。结果梅毒螺旋体接种组其形态学变化显示接种后第9天兔睾丸开始肿胀变硬,12~15d肿胀程度最为明显,之后肿胀程度及硬度逐渐降低;病理学变化显示接种梅毒螺旋体后3d家兔睾丸组织出现多形核白细胞和淋巴细胞浸润;7d淋巴细胞增多,多形核白细胞减少;14d有明显的单核细胞和淋巴细胞浸润,生精小管萎缩,精子产生减少;21d兔睾丸中淋巴细胞充满睾丸间质,生精管萎缩明显,成熟精子显著减少。兔睾丸镀银染色结果显示3d组梅毒螺旋体位于生精小管中,7d组及14d组显示梅毒螺旋体位于间质区,而在21d组的兔睾丸中镀银染色未发现梅毒螺旋体。结论梅毒螺旋体感染后家兔睾丸炎症反应逐渐加重,生精小管萎缩,精子数量明显减少,这可能与梅毒螺旋体诱发宿主免疫反应的产生和它本身入侵所造成的组织损伤有关。
Objective To investigate the effects of Treponema pallidum on pathological damage of testis and the sperms in New Zealand rabbit Methods The testes of 8 and 4 New Zealand rabbits aged 3 months were inoculated with Treponema pallidum (Nichols strain) and its inactivated strains respectively. After inoculation, the morphology ofNew Zealand rabbit testes was observed regularly, and the rabbit testes were obtained at different time for observing, the pathological and spenn changes. Results Morphological changes of the testicle exhibited swell and harden on the 9 th day after inoculation, and obvious swollen obvious on the 12th to 15th day, after which, the swollen and hardness gradually decreased. Meanwhile, polynuclear leukocyte and lymphocyte infiltration appeared in testicular tissues of the rabbits on day 3 after inoculation with Treponema pallidus. Lymphocytosis and polymorphonuclear leucopenia on day 7;On day 14,there were obvious infiltration of monocytes and lymphocytes, atrophic spermatogenesis and decreased sperm production. On the 21st day, lymphocytes filled the testicular stroma, spermatogenesis atrophy was obvious, and mature sperm decreased significantly. Warthin-Starry silver staining of testicles in rabbits showed that Treponema pallidum was located in spermatogenic tubule in the 3-day group, interstitial region in the 7-day group and the 14-day group, and no Treponema pallidum was found in the 21-day group. Conclusion After infected with Treponema pallidum, testicular inflammation gradually aggravated, spermatogenesis atrophic, and the number of sperm significantly reduced, which may be related to Treponema pallidum-induced host immune response and its invasion-caused tissue damage.
