肾癌组织中原钙黏蛋白-10甲基化状态及临床意义 被引量:2

Methylation status of PCDH10 gene in renal cell carcinoma and its clinical significance

摘要 目的探讨肾癌组织中原钙黏蛋白-10(PCDH10)甲基化状态及临床意义。方法收集78例肾癌患者的癌组织标本及其中38例癌旁正常肾组织标本;采用甲基化特异性PCR法检测组织PCDH10甲基化情况,分析肾癌组织PCDH10甲基化状态与患者临床特征及预后的关系。结果肾癌组织PCDH10甲基化阳性率(73.1%)明显高于癌旁正常组织(21.1%),差异有统计学意义(P<0.05)。肾癌组织PCDH10甲基化阳性与患者性别、年龄无关,差异均无统计学意义(均P>0.05);与肿瘤T分期、Fuhrman分级、淋巴结转移有关,差异均有统计学意义(均P<0.05)。肾癌患者PCDH10甲基化阳性组5年总生存率(73.7%)明显低于甲基化阴性组(95.2%),差异有统计学意义(P<0.05)。结论肾癌组织PCDH10甲基化阳性率较高,与肾癌的发生、发展及患者预后不良相关。 Objective To investigate the methylation status of procadherin-10(PCDH10)gene in renal cell carcinoma and its clinical significance.Methods Seventy eight specimens of cancer tissue and 38 specimens of pericancerous renal tissues were collected from patients with renal cell carcinoma.Methylation specific PCR was used to detect the methylation status of PCDH10 gene in renal cell carcinoma and pericancerous renal tissues.The relationship of PCDH10 mentylation with clinicopathological features and prognosis of patients was analyzed.Results The methylation rate of PCDH10 in renal cell carcinoma tissues was significantly higher than that in adjacent tissues(73.1%vs 21.1%,P<0.05).PCDH10 methylation in renal cell carcinoma tissues was not correlated to gender and age of patients(P>0.05),but correlated with T stage,Fuhrman grade and lymph node metastasis(all P<0.05).The 5-year overall survival rate in patients with PCDH10 methylation was significantly lower than that in non-methylation group(73.7%vs 95.2%,P<0.05).Conclusion The methylation rate of PCDH10 in renal cell carcinoma is higher,which is related to the occurrence,development and poor prognosis of renal cell carcinoma.
