摘要
为益母草微粉饲料原料的质量控制提供参考,采用薄层色谱法(TLC)进行定性鉴别,激光粒度分析仪对样品进行粒径检测,高效液相色谱法(HPLC)测定益母草碱的含量。结果表明:在薄层图谱中,供试品在对照品斑点位置出现相应斑点,且无干扰,TLC可用于益母草微粉的鉴别;益母草微粉粒径的上限为小于48μm颗粒百分比不低于95%,下限为大于75μm颗粒百分比不超过1.0%。HPLC检测中,益母草碱的进样量为0.15~0.60μg时,进样量与峰面积线性关系良好(R=0.9996);10批样品的益母草碱含量变幅为0.073%~0.081%,平均为0.077%。建议控制益母草饲料原料中益母草微粉含益母草碱不少于0.60mg/g。
In order to provide references for quality control of Leonurus heterophyllus micropowder feed raw materials,TLC was used for qualitative identification,laser particle size analyzer was used for particle size detection,and high performance liquid chromatography(HPLC)was used for the determination of L.heterophyllus alkaloids.Results:The TLC images showed that the speckles appeared in the control speckles,and there was no interference,TLC could be used to identify L.heterophyllus micropowder.The upper limit of the particle size of L.heterophyllus micropowder was less than 48μm,the percentage of particles was no less than 95%,and the lower limit was more than 75μm,and the percentage of particles was no more than 1.0%.In the HPLC determination,the relationship between the amount of L.heterophyllus alkaloids and the peak area was good(R=0.999 6)when the amount of L.heterophyllus alkaloids was 0.15-0.60μg.The content of L.heterophyllus alkaloids in ten batches of samples was 0.073%-0.081%,and the average value was 0.077%.Therefore,the content of L.heterophyllus alkaloids should be no less than 0.60 mg per gram in L.heterophyllus micropowder feed raw materials.
