摘要
针对酶法合成熊去氧胆酸中蛋白质含量的测定,研究了4种通用方法(考马斯亮蓝染色法、双缩脲法、紫外吸收法、Folin-酚法)的适用性,其中使用考马斯亮蓝染色法测定熊去氧胆酸中蛋白质含量RSD最小,为0.25%。同时通过考察考马斯亮蓝染色法的精密度、稳定性和重现性等参数,表明其稳定性较好,RSD为0.809%,重现性实验中的RSD为0.974%,专属性与耐用性均良好,回收率在98.33%-101.00%之间,准确度较高。
A comparative study was conducted in the protein detection of ursodeoxycholic acid during the following methods:coomassie brilliant blue staining method,biuret method,ultraviolet absorption method,Folin-phenol method.Results showed that coomassie brilliant blue staining method was the most accurate one in which the RSD was 0.25%.The precision,stability and reproducibility of coomassie brilliant blue staining method were also detected.This method showed good stability,specificity and durability with 0.809%in RSD,0.974%in reproducibility RSD and 98.33%-101.00%in recovery rate.
