摘要
目的:探讨人结肠癌细胞中miR-145-5p、miR-143-3p的表达发生变化后对P-gp和MRP的影响。方法:采用Western Blot和qRT-PCR法检测人结肠癌细胞分别转染miR-145-5p mimics/inhibitor、miR-143-3p mimics/inhibitor后P-gp和MRP的表达。结果:转染miR-145-5p mimics后P-gp和MRP的蛋白和基因表达均降低(P<0.05),转染miR-145-5p inhibitor后P-gp和MRP的蛋白和基因表达均升高(P<0.05)。转染miR-145-5p mimics NC和miR-145-5p inhibitor NC后P-gp和MRP的蛋白和基因表达无明显变化(P>0.05)。转染miR-143-3p mimics后P-gp和MRP的蛋白和基因表达均降低(P<0.05),miR-143-3p inhibitor后P-gp和MRP的蛋白和基因表达均升高(P<0.05)。转染miR-143-3p mimics NC和miR-143-3p inhibitor NC后P-gp和MRP的蛋白和基因表达无明显变化(P>0.05)。结论:miR-145-5p及miR-143-3p对多药耐药蛋白P-gp、MRP具有调控作用,参与了结肠癌的多药耐药。
Objective: To investigate the effect of the expression changes of miR-145-5 p and miR-143-3 p on P-gp and MRP in colon cancer cells. Methods: The expressions of P-gp and MRP in colon cancer cells transfected with miR-145-5 p mimics/inhibitor and miR-143-3 p mimics/inhibitor were detected by Western Blot and qRT-PCR. Results: Protein and gene expression of P-gp and MRP were decreased after transfection with miR-145-5 p mimics( P < 0. 05),and increased after transfection with miR-145-5 p inhibitor( P < 0. 05). Protein and gene expression of P-gp and MRP were not significantly changed after transfection with miR-145-5 p mimics NC and miR-145-5 p inhibitor NC( P > 0. 05). Protein and gene expression of P-gp and MRP were decreased after transfection with miR-143-3 p mimics( P < 0. 05),while protein and gene expression of P-gp and MRP were increased after transfection with miR-143-3 p inhibitor( P < 0. 05). Protein and gene expression of P-gp and MRP showed no significant changes after transfection with miR-143-3 p mimics NC and miR-143-3 p inhibitor NC( P> 0. 05). Conclusion:miR-145-5 p and miR-143-3 p have regulatory effects on multidrug resistant proteins P-gp and MRP,and are involved in the occurrence,development and multidrug resistance of colon cancer.
