miR-195及其靶向蛋白CHEK1与非小细胞肺癌预后的关系

Relationship between miR-195,target protein CHEK1 and prognosis of NSCLC

目的探讨非小细胞肺癌(NSCLC)组织中miR-195及其靶向蛋白CHEK1表达与预后的关系。方法采用qRT-PCR方法检测miR-195在组织标本中的表达水平;采用Western blot检测miR-195的靶向蛋白CHEK1的表达,比较患者NSCLC组织和正常肺组织中miR-195及其靶向蛋白CHEK1表达差异,并分析miR-195及其靶向蛋白CHEK1表达与NSCLC患者临床病理特征的关系。结果NSCLC组织中miR-195相对表达量较正常组织降低,靶向蛋白CHEK1表达量较正常组织升高(t分别=18.10、41.97,P均<0.05);miR-195及其靶向蛋白CHEK1表达与疾病分期、淋巴结转移及远处转移相关,差异均有统计学意义(t分别=12.40、11.19;11.95、10.88;11.07、11.88,P均<0.05),不同性别、年龄、有无吸烟史患者之间的miR-195及其靶向蛋白CHEK1表达比较,差异均无统计学意义(t分别=1.97、0.78;0.56、0.02;1.90、1.60,P均>0.05)。结论miR-195及其靶向蛋白CHEK1表达在NSCLC患者中表达异常,并与患者临床病理特征相关,可能与患者的预后生存密切相关。

Objective To investigate the relationship between miR-195 and its target protein CHEK1 expression and prognosis in NSCLC tissues.Methods The expression level of miR-195 in tissue samples was detected by QRT-PCR.Western blot was used to detect the expression of miR-195’s targeted protein CHEK1.The differences in miR-195 and its targeted protein CHEK1 expression in NSCLC tissues and normal lung tissues were compared,and the relationship between miR-195 and its targeted protein CHEK1 expression and clinical characteristics of NSCLC patients were analyzed.Results The relative expression level of miR-195 in NSCLC tissues was lower than that in normal tissues(t=18.10,P<0.05),and the targeted protein CHEK1 was higher than that in normal tissues(t=41.97,P<0.05).The expression of miR-195 and its target protein CHEK1 were correlated with lymph node metastasis and distant metastasis of disease stage,and the differences were statistically significant(t=12.40,11.19;11.95,10.88;11.07,11.88,P<0.05).The expressions of miR-195 and its target protein CHEK1 were not statistically related to gender,age,and smoking history(t=1.97,0.78;0.56,0.02;1.90,1.60,P>0.05).Conclusion The expressions of miR-195 and its targeted protein CHEK1 are abnormal in NSCLC patients,and it is correlated with the clinical features of patients,which may be closely related to the prognosis and survival of patients.
