摘要
应用透射电镜观察组织细胞的超微结构是生物医学研究的重要手段。成功包埋样品是成功制备超薄切片和获得清晰图像的先决条件。分别使用Spon812和Spurr两种环氧树脂对某些特殊生物样品进行包埋,并与联合使用两种树脂包埋的效果进行了比较,结果显示,Spon812树脂包埋的样品可获得反差显著的电镜图像,但对某些致密组织浸透不良,有空洞或褶皱形成;Spurr树脂包埋样品的浸透良好,尤其适合作为神经组织包埋剂,但图像反差较弱;联合使用两树脂包埋样品的浸透良好,图像反差介于两者之间。因此,联合使用两种树脂可以综合它们的优点,适合绝大多数生物样品的批量包埋。
Epoxy resins Spon 812 and Spurr are two common embedding medium on the TEM samples.The comparison result shows that the ultrastructural images of specimens embedded into Spon812 resin appear with high contrast,but the resin mixture penetration is not completed on some special tissues and some small cavities and folds are found on ultrathin sections.The specimens embedded into Spurr is highly efficient for penetration and appropriate for nerve tissues,but images contrast is lower than that of Spon812 resin.When Spon 812 and Spurr are combined used to embed specimens,the result is satisfied with good resin penetration and proper images contrast.This method is suitable for most of TEM biological specimens.
