基于网络药理学的鱼腥草治疗支气管炎作用机制研究 被引量:11

摘要 目的运用网络药理学方法筛选鱼腥草治疗支气管炎的有效成分和作用靶点,探究其成分-靶点-疾病的关系。方法运用中药系统药理学数据库与分析平台和文献检索收集鱼腥草化学成分并筛选活性成分,多个数据库联用检索查询成分和疾病对应的靶点,并利用Cytoscape3.5.1软件构建成分靶点-疾病靶点的可视化网络拓扑关系图,对成分靶点与疾病靶点进行可视化分析,通过基因功能富集平台GATHER分析靶点功能及机制通路。结果筛选出鱼腥草活性成分12个,得到可作用于支气管炎靶点42个。根据可视化网络拓扑图信息,得到关于鱼腥草治疗支气管炎5个核心靶点CD19、CD79A、TNFRSF13C、NFKB1、EGFR。GATHER筛选结果提示,鱼腥草的关键靶标富集到γ-六氯环己烷降解、脂肪酸代谢、补体和凝血级联等8条通路。结论鱼腥草可能主要通过抗病反应、免疫反应、生物刺激反应等发挥治疗支气管炎的作用。 Objective To identify the active constituents and targets of Houttuyniae Herba on bonchitis using the method of network pharmacology;To explore the relationship among“component-target-disease”.Methods TCM Systems Pharmacology Database and Analysis Platform was used and related literature was collected to screen out the active constituents of Houttuyniae Herba.Multiple databases were used to search the targets of the constituents and the disease,and Cytoscape3.5.1 software was used to construct a visual network topological relationship map of the target-disease target,and the constituent targets and the disease targets were visually analyzed.The target function and mechanism pathway were determined through the gene function enrichment platform GATHER.Results Twelve active constituents of Houttuyniae Herba were screened out and 42 related targets were obtained which could act on bronchitis.From the visual network topology map,five core targets,including CD19,CD79A,TNFRSF13C,NFKB1 and EGFR of Houttuyniae Herba were found.The GATHER database suggested that key targets of Houttuyniae Herba were enriched into 8 pathways,includingγ-hexachlorocyclohexane degradation,fatty acid metabolism,complement and coagulation cascades.Conclusion Houttuyniae Herba mainly exerts its effects on bonchitis via anti-disease reaction,immune response and bio-stimulatory reaction.
