摘要
目的基于网络药理学方法探究葛根治疗冠心病的作用机制。方法通过化学专业数据库收集葛根化学成分并对其进行类药性分析,预测作用靶点,构建成分-靶点网络;通过CooLGeN检索疾病相关基因,与葛根作用靶点做交叉分析,发现葛根治疗靶点,并进行蛋白质相互作用网络分析;最后进行GO生物学过程富集分析、通路富集分析。结果葛根的成分-靶点网络包含70个成分、180个相应靶点,其中18个靶点为葛根治疗冠心病的靶点。主要生物过程包括氮和氧的代谢与合成及脂肪酸代谢,主要通路包括血小板活化通路、花生四烯酸代谢通路、AGE/RAGE通路、一氧化氮作用通路等。结论葛根通过多靶点、多通路发挥对冠心病的治疗作用,一氧化氮作用通路、AGE/RGAE通路、花生四烯酸通路等可能是葛根治疗冠心病的主要通路。
Objective To study the molecular mechanism of coronary disease treated by Puerariae Lobatae Radix based on network pharmacology.Methods Chemical components of Puerariae Lobatae Radix were collected through chemical professional database and the drug-like properties were analyzed;the targets were predicted;a component-target network was constructed.Disease-related genes were retrieved by CooLGeN and were cross-analyzed with the targets of Puerariae Lobatae Radix;the therapeutic targets of Puerariae Lobatae Radix were found;protein interaction network analysis was performed.Finally,the enrichment analysis of GO biological process and pathway enrichment analysis were carried out.Results Component-target network of Puerariae Lobatae Radix contained 70 components and 180 corresponding targets,of which 18 targets were targets for the treatment of coronary heart disease.The main biological processes included the metabolism and synthesis of nitrogen and oxygen and fatty acid metabolism.The main pathways included platelet activation pathway,arachidonic acid metabolic pathway,AGE/RAGE pathway,and nitric oxide pathway.Conclusion The therapeutic effect of Puerariae Lobatae Radix on coronary disease may be realized through a multi-targets and multi-pathways way,and platelet activation pathway,arachidonic acid metabolic pathway,and AGE/RAGE pathway may play an important role in coronary disease treated by Puerariae Lobatae Radix.
